摘  要:探讨Bcl-2和Bax在C57/BL6小鼠海马发育和老化过程中的表达变化。方法:取胚龄(E)18、20d和生后(P)1、3、5、7、14、21、28d以及2、3、6、15、18个月(M)的CA7/BL6小鼠海马,应用免疫组织化学、免疫印迹法及图像分析技术检测Bcl-2和Bax蛋白的表达。结果:E18d~P7d,海马CA区及齿状回Bcl-2和Bax阳性细胞逐渐密集。P14-P21d,CA区两种阳性细胞稀疏;齿状回Bcl-2阳性细胞逐渐稀疏,Bax阳性细胞P14d密集,P21d稀疏。P28d后均趋于稳定。图像分析显示,E18d-P21d,除12)(3区Bcl-2的平均光密度值无差异外,BcP2和Pax均表现为先增大后降低的趋势。免疫印迹法结果显示,Bcl-2和Bax表达E18d~P7d逐渐增多,P14~P21d减少;P28d后趋于稳定。Bcl-2/Bax在P1d降低,P3d~P5d增高,P7d~P21d降低,P28d后趋于稳定。结论:Bcl-2和Bax在小鼠海马发育早期表达较多,成年及老年期处于较低水平,表明它们可能参与了海马的塑形过程。To observe the changes of expression of Bcl-2 and Bax protein in the development and aging of C57/ BL6 mouse hippocampus. Methods: Embyronic (E) 18, 20 day (d) and postnatal 1, 3, 5, 7, 14, 28 d and 2, 3, 6, 15, 18 month (M) mice were used in this study. Each group included 8 mice. Immunohistochemistry and immunoblot asssay were used to detect the expression of Bcl-2 and Bax protein in the mice hippocampus of different age. Results: From El6 d to P7 d, Bcl-2 and Bax immunoreactive cells increased in the hippocampus. On P14 d-P21 d, the two kinds of immunoreactive cells decreased, except the Bax positive cells in dentate gyrus which increased on P14 d while decreased on P21 d. Then this two kinds of cells remained unchanged after P28 d. Additionally, the average optical density of Bcl-2 and Bax on El8 d-P21 d in- creased firstly and then decreased, except the Bcl-2 in dentate gyrus area. Immunoblotting analysis showed that the expres- sion of Bcl-2 and Bax protein increased on E18 d-P7 d, decreased on P14 d-P21 d, and remained unchanged after P28 d. Bcl- 2/Bax decreased on P1 d, increased on P3 d-P5 d, decreased on P7 d-P21 d, and remained unchanged on P28 d. Conclusion: The expression of Bcl-2 and Bax is high in the early stage of hippocampus development and then low in adult and senectitude. It may be related with the formation of the hippocampus.
