摘 要:目的:通过建立鼠肺炎支原体肺炎模型,研究CdSe/ZnS量子点标记技术检测肺炎支原体抗原的最佳时期,并探讨其临床应用的可行性。方法:以CdSe/ZnS量子点标记兔抗肺炎支原体P1重组蛋白IgG作为检测抗体,对不同感染时期的鼠支气管灌洗液标本及105份临床呼吸道感染标本进行测定,观察CdSe/ZnS量子点标记方法检测肺炎支原体感染阳性率与病程的关系,以及该方法与聚合酶链反应(PCR)检测法的符合率。结果:感染3 d~7 d的鼠支气管灌洗液检测Mp阳性率最高。临床标本PCR检测阳性率为35.2%(37/105),量子点标记方法阳性率为31.4%(33/105),两者符合率达86.7%(91/105)。结论:量子点标记方法适用于肺炎支原体感染早期的检测,方法操作简单、检测快速,在实验室诊断中有良好的应用前景。Objective:Through the establishment of model mice infected by Mp, to explore the optimum time for antibody - quantum dots method in detection of Mycoplasma pneumoniae(Mp) antigen and evaluate its value for clinical application. Meth- ods: CdSe/ZnS quantum dots was used to make detection antibody by labeling rabbit anti - Mycoplasma pneumoniae P1 recombi- nant protein IgG in the bronchial lavage fluid of model mice in different infected periods and clinical respiratory tract infection samples. 105 clinical samples and bronchial lavage fluid specimens were examined by quantumdots(QDs) to determine the rela- tionship of Mp positive rate and disease progression, and to observe coincidence rate between the results of QDs and PCR meth- ods. Results: QDs for M. pneumoniae indicated highest positive rate was found after infection incidence in 3 ~ 7 days. In the 105 clinical samples, 37 were positive detected by PCR method and 33 were positive detected by the newly established method. The coincidence rate of these two methods was 86.7%. Conclusion: Results of this study indicate that quantum dot labeling method is an easy and rapid test for early detection of Mycoplasma pneumoniae infection, which may offer a good prospect in laboratory di- agnosis.
