摘 要:目的:观察microRNA-150与胶原Ⅰ在糖尿病心肌纤维化大鼠和心肌成纤维细胞中的表达,探讨microRNA-150在糖尿病心肌纤维化中对胶原Ⅰ的影响。方法:制备糖尿病大鼠模型,原代培养心肌成纤维细胞。masson染色观察心肌形态、胶原容量;Elisa法检测心肌组织和心肌成纤维细胞中胶原Ⅰ的表达;Real-time PCR检测心肌组织和心肌成纤维细胞中microRNA-150的表达。结果:糖尿病组大鼠心肌内胶原容量明显高于对照组(P<0.05),在糖尿病组大鼠和高糖培养的心肌成纤维细胞中胶原Ⅰ分泌增加(P<0.05),microRNA-150的表达降低(P<0.05);细胞转染microR-NA-150 micic后,高糖培养的心肌成纤维细胞胶原Ⅰ分泌减低(P<0.05)。结论:microRNA-150在糖尿病心肌纤维化中可影响胶原Ⅰ的分泌,提示microRNA-150可能参与糖尿病心肌纤维化的发生。Objective:To observe the expression level of microRNA - 150 and collagen I on myocardial fibrosis in diabetic rat model and myocardial fibroblasts, to discuss the influence of microRNA - 150 on collagen I in diabetes myocardial fibrosis. Methods: The diabetic rats were induced. The myocardial fibroblast were primary cultured. Masson staining and Elisa were used to detect collagen I ~ Real -time PCR was used to detect microRNA - 150. Results: The collagen I level was higher in the DM group. The expression of microRNA- 150 was downregulated in the DM group; Collagen I of myocardial fibroblasts which were cultured in high glucose were downregulated after myocardial fibroblasts were transfected microRNA - 150 micic. Conclusion: The microRNA -150 level can influence Collagen I secretion in diabetes myocardial fibrosis, microRNA -150 might be involved in diabeties myocardial fibrosis.
