摘  要:目的:观察参元丹含药血清对缺氧复氧心肌细胞自噬的影响,探讨参元丹心肌保护作用机制。方法:建立缺氧复氧心肌细胞损伤模型,采用免疫细胞化学法观察心肌细胞微管相关蛋白1轻链3(LC3)和磷酸肌醇-3-激酶(PI3K)表达,实时荧光定量PCR(Real-time PCR)法测定自噬相关基因(Beclin-1)和半胱天冬氨酸蛋白酶-3(Caspase-3)mRNA表达量。结果:与正常对照组相比,缺氧复氧组LC3和PI3K荧光表达增强,参元丹含药血清组与相应浓度空白血清组相比,细胞LC3和PI3K荧光表达强度减弱,其中参元丹含药血清10%组PI3K荧光强度明显降低,具有统计学意义(P<0.05);缺氧复氧组细胞Beclin-1和Caspase-3 mRNA表达量明显升高,与相应浓度空白血清组相比,参元丹含药血清组Beclin-1 mRNA表达水平明显降低;参元丹含药血清5%组Caspase-3 mRNA表达量降低。结论:参元丹含药血清对缺氧复氧细胞自噬具有抑制作用,可能与抑制PI3K,下调Beclin-1和Caspase-3基因有关。Objective: To study the influence of medicated serum with Shenyuan Dan(SYD) on autophagy of cardiac myocyte induced by hypoxia/reoxygenation(H/R),and investigate its myocardial protective mechanism.Methods: To establish myocardial H/R injury model.The expressions of LC3 and PI3K in cardiac myocyte were observed respectively using immunocytochemistry.The mRNA expressions of Beclin-1 and Caspase-3 in cell were determined using real-time PCR.Results: Compared with control group,both the fluorescence expression of LC3 and PI3K in I/R group increased,while the expression of LC3 in medicated serum with SYD group decreased,and the expression of PI3K in 10% medicated serum with SYD group obviously decreased and showed statistically significance(P〈0.05);The mRNA expression of Beclin-1 and Caspase-3 in I/R group increased.Medicated serum group,compared with control serum group,the mRNA expression of Beclin-1 decreased,and that of Caspase-3 in 5% group decreased.Conclusion: Medicated serum with SYD had inhibition on cardiomyocytes autophagy induced by H/R.The mechanism might be related to restrain PI3K,and down-regulate the gene of Beclin-1 and Caspase-3.
