摘 要:目的:观察扶正抗病毒方对A型流感病毒感染小鼠肺部病理变化与超微结构的影响。方法:采用鼻腔接种法建立A型流感病毒小鼠肺部感染模型。实验分为正常组、模型组、利巴韦林组和扶正抗病毒方组,经灌胃给药或给0.9%氯化钠溶液7d后,处理动物,常规检测肺重量指数;光学显微镜技术观察肺病理变化,透射电子显微镜技术观察肺组织超微结构。结果:①与正常组比较,模型组肺指数明显升高(P<0.01),扶正抗病毒方组肺指数降低,与模型组比较,差异显著(P<0.05)。②与正常组比较,模型组肺脏充血、水胀,呈现暗褐色外观,肺泡、细支气管等正常组织结构消失,肺泡内有大量炎症细胞浸润。扶正抗病毒方组肺组织病理变化显著改善。③与正常组比较,模型组小鼠肺组织超微结构受到破坏,核膜与质膜模糊不清,细胞器减少,线粒体嵴减少或消失,内质网出现空泡样病变,胞浆内有大量病毒颗粒。扶正抗病毒方组肺组织细胞结构较完整,细胞器丰富,线粒体和内质网的改变程度低于模型组。结论:扶正抗病毒方对A型流感病毒感染所致的肺部病理变化与超微结构有改善作用。Objective: Observing the effect of SACR on pulmonary pathological change and ultrastructure of mice infected by influenza virus type A.Methods: Use nasal inoculation to establish mouse pulmonary infection model animals by A-type influenza virus.The animals were divided into normal group,model group,ribavirin group and SACR group.After 7 days of lavaging drug or normal saline,dispose mice and detect pulmonary weight index routine;Observing pulmonary pathological change and ultrastructure by light microscope and transmission electron microscope.Results: ①Compared with normal group,pulmonary index of model group is significantly increased(P〈0.01).Pulmonary index of SACR group is decreased,which is different from model group obviously(P〈0.05).②Compared with mormal group,lungs of model group are hyperemic and dropsical,and showing dust-color,normal organizational structure such as alveolus,bronchiole are disappeared,mass inflammatory corpuscle infiltrate within lungs.The pathological change of lungimproved significantly in SACR group.③Compared with normal group,pulmonary organization structures are destroyed in model group,karyolemma and karyoplasm are inexplicit,the quantity of cell organs and mitochondrial cristas become smaller or even disappear,vacuolated changes in endoplasmic reticulum show up and there are plenty of viral particles in endochylema.In SACR group,pulmonary organizational structure nearly intact,cell organ plentiful,degree of mitochondria and endoplasmic changed lower than model group.Conclusion: SACR play a part in mending pulmonary pathological change and ultrastructure of mice infected by A-type influenza virus.
