摘 要:目的:探讨银花合剂对急性肺损伤(ALI)大鼠干预作用与可能机制。方法:采用油酸加内毒素"二次打击"方法复制大鼠ALI模型,用银花合剂进行干预,实验结束后测定动脉血氧分压、肺湿/干质量比值、肺病理形态学积分、血浆IL-1β的含量及大鼠海马CA2-3区FOS蛋白阳性细胞灰度值。结果:与对照组比较,模型组大鼠IL-1β含量显著升高(P<0.01),海马FOS阳性细胞灰度值迅速下降(P<0.01),与模型组相比,银花合剂组和地塞米松组氧分压上升,肺湿/干质量比值、病理形态学积分、IL-1β含量显著下降(P<0.01),FOS蛋白阳性细胞灰度值明显增高(P<0.01)。结论:银花合剂能够减轻肺损伤,其可能机制是银花合剂通过拮抗IL-1β,抑制海马c-fos表达实现对急性肺损伤大鼠的防治作用。Objective: To study the effects and possible mechanisms of Yinhua Agent on the acute lung injury(ALI)rats.Methods: To build the’Two hit’animal model of acute lung injure in rats induced with oleicacid and lipopolysaccharide.The levels of arterial partial pressure of oxygen,wet-to-dry weight ratio of lung,pathomorphological score,plasma IL-1β,gray value of fos positive cell in CA2-3 regoin of rats hippocampus were detected.Results: In comparison with normal group the level of plasma IL-1β tended to elevate significantly(P〈0.01)and the gray values of fos positive cell declined dramatically in model group(P〈0.01).But the PaO2 was improved and the level of ratio W/D,pathomorphological score,plasma IL-1β were decreased significantly in Yinhua Agent and DEX groups compared with model group(P〈0.01);the gray values in Yinhua Agent and DEX groups increased significantly(P〈0.01).Conclusion: The results suggested that Yinhua Agent played a protective role in ALI.The mechanism of effects might be inhibition of the content of plasma IL-1β and the expression of hippocampus c-fos gene.
