摘 要:目的:探讨活血、利水中药对脑出血大鼠蛋白酶激活受体-1(PAR-1)表达的影响。方法:168只雄性SD大鼠随机分为假手术组、模型组、活血组和利水组。采用自体不凝血注入尾状核法制作大鼠脑出血模型,分别于术后1、3、5d 3个时间点取材,组织干湿重法计算脑含水量,免疫组织化学法和Real-time PCR法检测脑组织PAR-1蛋白和基因的表达。结果:与假手术组相比,模型组各时间点大鼠脑组织含水量增多(P<0.01),PAR-1蛋白和基因的表达均明显升高(P<0.01);活血组和利水组大鼠脑组织含水量及PAR-1的表达较模型组降低(P<0.05),且活血方优于利水方(P<0.05)。结论:活血、利水中药均能减轻脑出血后脑水肿,其机制可能与降低PAR-1表达有关。Objective: To investigate the effects of activating blood and diuresis traditional Chinese medicines on changes of cerebral edema and expression of PAR-1 within intracerebral hemorrhage(ICH) rats.Method: 120 SD rats were randomly divided into the following groups: the sham operated group,model group,activating blood group and diuresis group,30 rats in each group.Experimental intracerebral hemorrhage rat model was established by injection autologous blood into caudate nucleus,rats were sacrificed at three time points of 1 day,3 days,5 days after surgery respectively,10 rats in each time.The brain water content was determined by tissue wet-dry weight method,the expression of PAR-1 protein and PAR-1mRNA in cerebral tissues was determined respectively by immunohistochemitry techniques and real-time PCR.Result: Compared with the sham group,the expression of PAR-1 mRNA and PAR-1 protein increased significantly(P〈0.01) in the model group,and the brain water content had the same trend(P〈0.01);While at the same time,compared with the model group,the expression of PAR-1 mRNA and PAR-1 protein and the brain water content decreased obviously in activating blood group and diuresis group(P〈0.05),there were also significances in statistics between the activating blood group and diuresis group(P〈0.05).Conclusions: Activating blood and diuresis traditional Chinese medicines can decrease the edema of ICH,the probable mechanism is that it could regulate the expression of PAR-1.Furthermore,activating blood traditional Chinese medicines are better than diuresis traditional Chinese medicines.
