摘 要:目的利用不同方法对荭草DNA提取,比较提取质量的差异,为研究药材的遗传多样性、种类鉴定和基因指纹图谱的构建等提供优质基础。方法采用改进的CTAB法和SDS法提取荭草叶片基因组DNA,利用琼脂糖凝胶电泳、紫外分光光度法测定样品OD260/OD280的比值,检测所提DNA的纯度和质量。结果采用CTAB法提取的荭草基因组DNA样品的纯度和浓度均高于SDS法,其DNA的OD260/OD280值为1.801~1.993,CTAB法所提的DNA泳带分布较集中,无拖尾现象,其DNA浓度能够满足RAPD、SSR/ISSR扩增模板的需求。结论改良CTAB法是提取荭草基因组DNA的最佳方法。Objective To study the extraction methods of genomie DNA from the leaves of Polygonum orientale L. and to provide the basic guarantee for studying the genetic diversion, the species identification and the construction of gene fingerprint and so on. Methods With leaves of Polygonum orientale L. as tested materials, the genornic DNA was extracted using the modified CTAB method and SDS method. Results The DNA purity and concentration of samples extracted with CTAB method were higher than those with the SDS method. The ratio of OD260/OD280 was 1. 801 - 1. 993. DNA bands extracted with CTAB method were centralized distribution and non - tailing phenomenon which could meet the requirements of RAPD, SSRVISSR amplification template. Conclusion It is effective to extract the genomic DNA from Polygonum Orientale L. by modified CTAB method.
