摘 要:目的应用高速逆流色谱(HSCCC)分离人参中的人参皂苷,并鉴定分离所得到的化合物。方法以溶剂配比为醋酸乙酯-正丁醇-水-醋酸(4∶1∶3∶0.02,V∶V∶V∶V)作为制备型逆流色谱分离的溶剂系统,以上相为固定相,下相为流动相,流速为1.5 ml.min-1,仪器转速为950 r.min-1,检测波长254 nm,并利用高效液相色谱对分离所得到的组分与标准品进行对照,确定单体皂苷。结果应用高速逆流色谱技术一次性分离得到Re、Rg1、Rg33个人参皂苷单体化合物,经高效液相色谱(HPLC)检测其纯度均达到95%以上。结论高速逆流色谱分离单体化合物具有迅速、简单、重复性好的特点。Objective To separate ginsenosides from ginseng by high speed counter current Chromatography (HSCCC) and identify by high performance liquid chromatography (HPLC). Methods The optimized two - phase solvent system of the HSCCC was ethyl acetatebutanol- water- acetic acid (4: t: 3: 0.02 , V/V/V/V). The lower phase was mobile phase and the upper phase was stationary phase, the flow rate was 1.5 ml. min^-1, the revolution speed was 950 r. min-X and the detection wavelength was set at 254 nm. The separated fractions were identified with standard samples by HPLC. Results The three ginsenosides were obtained from the ginseng by HSCCC, the contents of three ginsenoside Re, Rgl, Rg3 were above 95%. Conclusion The HSCCC method is a simple, effective and reproducible tool for the separation of the compounds from natural plants.
