Observation on the Analgesic Effect of Repeated Electroacupuneture and Its Relation to the Expression of CAMKⅡ in Rat Hypothalamus

摘  要:目的观察电针(EA)镇痛的累积效应,及其与下丘脑CAMKⅡ表达的关系。方法 Wistar雌鼠70只,随机分为正常对照组(n=10),慢性压迫性损伤(CCI)组(n=30),去卵巢(OVX)+CCI组(n=30)。后两组又各分为不电针、EA 2天(2 d)和2周(2w)组,每组10例。OVX动物去除双侧卵巢,每天颈背部皮下注射D-半乳糖,7周后水迷宫法测试动物学习记忆能力。结扎坐骨神经造成CCI慢性痛模型。电针双侧"足三里"-"阳陵泉"穴,1次/d,不同组分别电针2 d、2W。辐射热照射测定大鼠缩腿潜伏期,以两足的差值作为痛敏分数(HAS)。用免疫组化法测定下丘脑室旁核(PVN)CAMKⅡ阳性产物的表达。结果 CCI后,动物HAS明显增大。CCI+A2W组HAS的绝对值显著低于CCI组、CCI+EA 2 d组(P<0.05);并且显著低于OVX+CCI+EA2W组(P<0.05)。与CCI组比较,CCI+EA2W组下丘脑PVN中CAMKⅡ积分灰度值明显较低(表达上调;P<0.05);而CCI+EA 2 d组无明显变化。OVX+CCI+EA2W组CAMKⅡ积分灰度值明显低于OVX+CCI组(P<0.05),而显著高于CCI+EA2W组(P<0.05)。与CCI+EA2W组比较,OVX+CCI+EA2W组CAMKⅡ表达的上调程度低,有显著性意义(P<0.05)。结论重复电针有累积镇痛效应;下丘脑CAMKⅡ表达上调与针刺累积效应密切相关;神经记忆力的减退在一定程度上减弱针刺镇痛的累积效应。Objective To observe the accumulative analgesic effect of electroacupuncture(EA) and its relation to the expression of Calcium/calmodulin-dependent protein kinaseⅡ(CAMKⅡ) in rat hippocampus and hypothalamus tissues in chronic constrictive injury(CCI) and ovariectomy(OVX) rats. Methods A total of 70 Wistar rats were randomized into control,CCI,CCI+EA 2 day(D),CCI+EA 2 week(W),OVX+CCI,OVX+CCI+EA2D and OVX+CCI+EA2W groups,10 cases in each group.For rats of the latest 3 groups,subcutaneous injection of D-galactose was given once daily for 5 w.Morris water maze test was conducted to evaluate the memory-learning ability.Chronic pain model was established by ligating the left sciatic nerve with a piece of catgut.EA(2/15Hz,1-2 mA) was applied to bilateral "Zusanli"(ST 36)-"Yanglingquan"(GB 34),for 30min,once daily for 2 d,2 w respectively.Radiant heat irradiation-induced paw withdrawal latency(PWL,pain threshold) was detected and PWL difference values between two paws were used as the hyperalgesia score(HAS).CAMKⅡ expression in paraventricular nucleus(PVN) of hypothalamus was detected by immunohistochemistry. Results Compared with control group,HAS was increased significantly after CCI.HAS in CCI+EA2W group was markedly lower than that of CCI and CCI+EA2D groups(P0.05).In comparison with OVX+ CCI +EA2W group,HAS in OVX+CCI+EA2W group was significant(P0.05).Interga(grey value igu of)CAMKⅡ immunoreaction positive product increased in rat hypothalamic PVN of CCI+EA2W group was significantly lower than that of CCI group(P0.05).IAV of CAMKⅡ is hypothalamus of OVX+CCI+EA2W group was significantly lower than that of OVX+CCI group(P0.05) but obviously higher than that of CCI+EA2W group(P0.05).
