摘　　要：目的:采用高效液相-蒸发光散射检测法(HPLC-ELSD)测定黄芪药材中黄芪甲苷的含量。方法:采用Venusil XBP C18(250.0 mm×4.6 mm,5μm)色谱柱,流动相:乙腈-水(3268),流速:1.0 mL.min-1,ELSD参数:雾化温度36℃,漂移管温度:60℃,通气压力30 psi。结果:黄芪甲苷在1.402 8～7.064 0 mg.L-1范围内有良好的线性关系,r=0.999 8(n=5),平均回收率为99.3%。结论:该方法分离度高,重现性好,灵敏度高,准确,简便,无干扰等优点,可广泛用于黄芪药材的质量控制。Objective:To determine astragaloside Ⅳ in Astragalus by HPLC-ELSD.Methods:HPLC-ELSD was used in the quantitative analysis of astragaloside Ⅳ in the sample.The separation was performed on Venusil XBP C18 column(25.0 mm×4.6 mm,5 μm,Agela Technologies) column with amixture of acetonitrile-water(3268) asmobile phase.The flow rate was 1.0 mL·min^-1.The temperature of drift tube formeasurement was 60 ℃.Results:The standard curve was linear in the range of 1.402 8~7.064 0 μg for astragalosideⅣ(r=0.999,n=5).The average recovery was 99.3%.Conclusion:The method is simple,reliable and good.It may be used for the quality control of astragali.
