薄膜超声法制备龙胆苦苷脂质体

摘　　要：目的制备均匀稳定的龙胆苦苷脂质体。方法采用薄膜超声法制备龙胆苦苷脂质体;采用HPLC法测定含量,色谱条件:流动相为甲醇:水=30:70,流速为1 mL/min,检测波长为270 nm;采用微柱离心法分离脂质体测量包封率。结果胆固醇:磷脂比1:2,PBS缓冲液的pH值为5.7,十八胺磷脂比1:10,药脂比为1:12。最优制备工艺采用薄膜超声法制备龙胆苦苷脂质体最佳包封率为75.14%。结论采用薄膜超声法制备龙胆苦苷脂质体包封率较好。Objective To prepare the liposomes Methods Preparing the liposomes of Gentiopicrin of Gentiopicrin by the film-ultrasonic dispersion method. by the film-ultrasonic dispersion method, using HPLC to determine the content of Gentiopicrin, and mini-column centrifugation was used to separate liposomes to determine the envelopment rate. Results The proportion of the cholesterol to phospholipid was 1：2,the pH value of buffer solution was 5.7, the proportion of the octadecyl amine to phospholipid was 1：10,the proportion of the drug to phospholipid was 1：12. The envelopment rate of the liposomes of Gentiopicrin prepared by the optimum process reached 75.14%.Conclusion The established method could prepare the liposomes of Gentiopicrin with good envelopment rate.
