摘 要:目的建立了苦豆子多糖含量的测定方法,确定了苦豆子多糖的含量。方法用苯酚-硫酸法,在波长490nm处测定吸光度,苦豆子多糖在20-100rμg/mL范围内具有良好的线性关系,相关系数为0.9995。平均回收率为101.4%,RSD=1.98%。结果苦豆子多糖的最佳提取条件为料液比1:10,提取2次,提取时间3h,苦豆子多糖换算因子为1.22,苦豆子中多糖的含量为90.90g/kg。结论该方法简便、准确率高、重现性好,可用于苦豆子中多糖含量的测定。Objective To establish an analysis method for the determination of polysaccharides from Sophora alopecuroides L.Methods The method of phenol-vitriol was used to determine the content of polysaccharide. The wavelength was set at 490 nm.The calibration curve of polysaccharide showed a good linearity within the range of 20-100 μg/mL. The average recovery was 101.4% with RSD=l.98%. Results The optimum extraction condition of polysaccharides was as follows: solid-liquid ratio was 1 : 10, extracted 2 times and each for 3 h. The converting factor was 1.22 and the content was 90.90g/kg.Conclusiou This method was simple, accurate and repeatable,which could be applied to determine the polysaccharides in Sophora alopecuroides L.
