摘  要:目的探讨重组大鼠白细胞介素-10(rIL-10)联合抗大鼠CD40L单克隆抗体对抑制大鼠小肠移植早期排斥反应的作用,为小肠移植免疫提供实验依据。方法将30只雄性SD大鼠和Wistar大鼠(各15只),建立(SD→Wister)大鼠小肠移植模型,随机分为A、B、C3组各10只。A组为生理盐水对照组;B组应用rIL-10;C组应用rIL-10联合抗CD40L单克隆抗体。3组均于术后第2天开始注射。术后第5、7天从3组随机各取5只,取小肠移植物,行组织病理学检查;采受体大鼠的外周血检测IL-2、干扰素-γ(INF-γ)表达水平。结果组织病理学检查:与A组相比,B、C组早期排斥反应明显延迟,且C组早期排斥反应较B组轻;外周血血清学检测:B、C组受体大鼠血清IL-2、INF-γ较A组明显降低,且C组较B组均有不同程度的下降,差异均有统计学意义(P<0.05)。结论 rIL-10联合抗大鼠CD40L单克隆抗体能有效抑制大鼠小肠移植的早期排斥反应。Objective To investigate the function of interleukin-10(rIL-10) combined with anti-CD40L monoclonal antibody in the early rejection of small intestinal transplantation on rat,and to provide experiment basis for intestinal transplantation immunity.Methods 30 male SD and Wister rats were selected as experimental subjects to establish(SD→Wister) rat small intestine transplantation model.All the experimental rats were randomly divided into 3 groups,ech of 10rats.A group was normal saline control group;B group injected rIL-10;C group injected rIL-10 combined with anti-CD40L monoclonal antibody.All injection began at the second days after surgery.On the 5th and 7th days after surgery,randomly selected 5 rats to acquire the small intestinal transplantations and then take examinations of histopathology;Drew peripheral blood to test the expression levels of IL-2,interferon-γ(INF-γ) .Results Histopathological examination:Compared with A group,early rejection in B,C group were significantly delayed,and early rejection in C group was lighter than B group;Blood serology:Serum IL-2,INF-γ of B,C group were significantly lower than those of A group,and C group compared with B group,was decreased in varying degree,the difference were statistically significant(P<0.05) .Conclusion rIL-10 combined with anti-rat CD40L monoclonal antibody can inhibit the rat small intestine transplantation in the early rejection.
