兔眼小梁切除术后滤过泡组织血管内皮生长因子和转化生长因子β2的相关性研究

摘　　要：目的观察兔眼小梁切除术后滤过泡瘢痕形成过程中血管内皮生长因子(vascular endothelial growth fac-tor,VEGF)与转化生长因子(transforming growth factor beta 2,TGF-β2)的表达变化及相关性,探讨VEGF和TGF-β2和在滤过泡瘢痕形成中的关系。方法随机选取24只青紫蓝兔双眼中的一眼作为实验组,行青光眼小梁切除术,另一眼不行手术作为正常对照组,另取2只兔不手术作为空白组。术前及术后第1、3、5、7、14、21天,观察滤过泡炎症反应和血管增生情况,采用双抗体夹心酶联免疫吸附法(enzyme-linked immunosorbent assay,ELISA)测量兔眼滤过泡组织中VEGF及TGF-β2的含量。结果与对照组比较,手术后滤过泡组织中VEGF和TGF-β2表达上调,分别于手术后第5、7天达高峰,差异有统计学意义(P<0.05),同一组织的VEGF和TGF F-β2含量有相关性(P<0.05)。结论青光眼小梁切除术后滤过泡组织内VEGF和TGF F-β2的含量明显增加,与瘢痕化进程一致,两者含量有相关性。Objective To observe the dynamic expression of vascular endothelial growth factor（VEGF） and transforming growth factor beta 2（TGF-β2） in the filtering bleb of rabbits after trabeculectomy,and determine their relationship.Methods Trabeculectomy was performed in one random eye of 24 chinchilla rabbits.The other eye was control.Another 2 rabbits were blank controls.At 1,3,5,7,14,21 days after operation,the inflammatory reaction and vascular proliferation status of the filtering bleb was observed,the VEGF and TGF-β2 contents were measured by enzyme-linked immunosorbent assay（ELISA）.Results Compared with the control group,the VEGF and TGF-β2 increased significantly and reached to peak at 5 and 7 days after operation respectively（P0.05）.VEGF and TGF-β2 contents had significant correlation（P0.05）.Conclusion VEGF and TGF-β2 contents in the filtering bleb are obviously increased,closely related with the scarring process and intercorrelated with each other.
