山茱萸总甙对实验性变态性脑脊髓炎大鼠外周一氧化氮的影响

摘  要:[目的]初步观察山茱萸总甙(TGCO)对实验性变态反应性脑脊髓炎(EAE)大鼠及外周血清一氧化氮(nitric oxide,NO)的影响。[方法]将Wistar大鼠分为3组:Control组、EAE组和TGCO组,采用动物行为学、常规HE和Luxol Fast Blue(LFB)染色观察动物的发病情况与中枢神经系统(CNS)的病理变化,硝酸还原酶法观察血清NO的变化。[结果]EAE组7只大鼠出现典型的EAE行为学改变、CNS炎性细胞浸润和髓鞘脱失,而TGCO组只有3只大鼠出现EAE行为学改变,评分最高为2分,潜伏期延长,CNS内炎性细胞浸润和髓鞘明显减轻。Control组未见上述变化。EAE组大鼠总血清NO值明显升高,与对照组比较有显著性差异(P<0.001);TGCO组大鼠血清NO值(109.83±68.03),与EAE组比较有统计学差异(P<0.05)。[结论]TGCO对EAE大鼠有抑制作用,可能与抑制血清NO合成有关。[Objective] To primarily investigate the effect of otal glycosides from cornus officinalis on nitric oxide of serum in experimental allergic encephalomyelitis in rats.[Methods] Wistar rats were randomly divided into three groups: Control group,EAE group,TGCC group.The diet,movement,body weight of Wistar rats were daily examined,CNS tissues were stained with Hematoxylin-eosin and Luxol Fast Blue,and nitric oxide in plasma were examined by nitric acid redox.[Result] Seven rats in EAE group demonstrated neurological signs of typical EAE,with parenchymal infiltration and perivascular cuffing with mononuclear cells,and demyelination in the white matter.However,three rats in TGCO group demonstrated neurological signs of typical EAE,the high score was 2,the latency was prolonged,the infiltration of mononuclear and myelin damages in CNS were reduced.No infiltration of mononuclear and myelin damages in CNS were noted in all rats of control groups.The level of NO in serum was(148.84±53.74)in EAE group obviously higher than that of Control group.The level of NO in serum was(109.83±68.03)in TGCO group significant lower than that of EAE Group.[Conclusion] TGCO could have an effect on prevention on EAE in Wistar rat,may be correlated with its ability to depress the synthesis of NO in serum.
