摘  要:目的:探究Aβ低分子量寡聚体对大鼠学习记忆功能的影响;方法:双侧海马内一次性注射Aβ低分子量寡聚体,15天后开始进行行为测试,测试方法采用跳台实验法、穿梭实验法和Morris水迷宫法;结果:跳台实验结果显示,与对照组比较,Aβ组跳台潜伏期短,错误次数多,差异有统计学意义(P<0.05);穿梭实验结果显示,与对照组比较,Aβ组由明箱进入暗箱的潜伏期短,错误次数多,差异有统计学意义(P<0.05);水迷宫实验结果显示,与对照组比较,Aβ组潜伏期长,跨越原平台次数少,差异有统计学意义(P<0.01);结论:Aβ组大鼠学习记忆能力和空间分辨能力降低,Aβ低分子量寡聚体在大鼠体内表现出较强的毒性作用。Objective: To investigate the effect of low molecular weight oligomers of β-amyloid peptide on the rat learning and memory.Methods: LMW Aβ oligomers was microinjected into bilateral hippocampus at once.And behavioral tests were carried out af-ter 15 days.Step-down test,Shuttle-box test and Morris water maze test were used to evaluate rat learning and memory function.Results: Step-down test showed that the rats in Aβ made notable errors compared with the control group(P0.05)and the incubation period(jumping from the platform) significantly reduced(P0.05).Shuttle-box test showed that compared with the control group,the rats in Aβ made notable errors(P0.05)and the incubation period(entering in dark chamber from light chamber) significantly reduced(P0.05).Morris water maze test indicated that the incubation period of finding platform significantly extended(P0.01) and frequency passing through the prior platform was less in Aβ group(P0.01).Conclusions: Microinjection of LMW Aβ oligomers into rat hip-pocampus may induce the learning and memory dysfunction,and LWM Aβ oligomers induced an impairment in rat spatial resolution.In a word,LWM Aβ oligomers had strong toxic effects on the rat learning and memory.
