摘  要:目的研究氯沙坦对肾间质纤维化大鼠肾组织ACE2 mRNA、TGF-β1 mRNA表达的影响。方法复制腺嘌呤致肾间质纤维化大鼠模型,随机分成氯沙坦组和模型对照组,氯沙坦治疗组大鼠予氯沙坦(10 mg·kg-1.d-1)混悬液灌胃30 d,模型组予等量蒸馏水灌胃,另设正常对照组予等量蒸馏水灌胃。在实验结束时留取血、尿、肾组织标本,检测Scr、BUN、24 h MTP等生化指标,采用Real Time PCR测定肾组织ACE2 mRNA、TGF-β1 mRNA表达水平。结果氯沙坦组较模型组Scr、BUN、24 h MTP均显著下降(P<0.01)。Real Time PCR结果显示模型组较正常组ACE2 mRNA表达水平下降(P<0.01),而TGF-β1 mRNA的表达水平反而增加(P<0.01);经治疗后氯沙坦组较模型组ACE2 mRNA表达水平明显增加(P<0.01),TGF-β1 mRNA的表达水平明显减少(P<0.01)。结论氯沙坦具有肾功能保护作用,其作用机制除了拮抗AT1R外,还可能与上调ACE2 mRNA表达水平,抑制TGF-β1 mRNA的表达水平而发挥抗肾间质纤维化作用有关。Objective To observe the effects of losartan on the expressions of ACE2 mRNA, TGF-β1 mRNA in renal tissues of chro- nic renal failure rats. Methods Rats were randomly divided into losartan and chronic renal failure groups after the Chronic renal failure rat model was prepared. Losartan group was received losartan suspension (10 mg · kg-1· d-1 ), and normal control , model groups received distilled water by gavage. The rats were sacrificed after 30 days' medication. Blood, urine and kidneys samples were collected to test kidney function, albuminuria and pathomorphology analysis. Real time PCR was used to detect the amounts of ACE2 mRNA and TGF-I31 mRNA ex- pressions. Results Compared with those of model group, the levels of Scr, BUN and MTP of losartan group were decreased significantly ( P 〈0. O1 ). Real time PCR results showed that ACE2 mRNA expression of model group was decreased significantly( P 〈0. 01 ), but TGF- 131 mRNA expression was increased(P 〈 0. 01 ). Compared with those of model group, ACE2 mRNA expression of losartan group was signifi- cantly increased (P 〈 0.01 ), but TGF-β1 mRNA expression was decreased ( P 〈 0. 01 ). Conclusions Losartan has renal protection, its mechanism may be connected with raising ACE2 mRNA expression, restraining TGF-β1 mRNA expression.
