摘 要:目的观察艾灸对抑郁症模型大鼠中缝核色氨酸羟化酶2基因表达(TPH2mRNA)以及血清和大脑皮质单胺氧化酶(MAO)活性的影响,探讨艾灸治疗抑郁症的作用机制。方法雌性sD大鼠,采用随机数字表将大鼠分为止常组(6只)、模型组(7只)和艾灸组(7只),孤养和慢性不可预见性温和应激(CUMS)制备抑郁症大鼠模型。利用RT—PCR技术检测大鼠中缝核TPH2mRNA的表达;比色法测试大鼠血清和大脑皮质MAO活力。结果艾灸组大鼠中缝核TPH2mRNA的相对表达量高于模型组,差异有统计学意义[相对密度值:(0.3759±0.0272),(0.2573±0.0581),P〈0.05];血清和大脑皮质MAO活力[分别为(1.919±1.243)U/ml,(1292.048±90.072)U/mg protein)]均低于模型组[分别为(4.117±2.727)U/ml,(1441.345±117.050)U/mg protein]。结论艾灸可能通过提高中缝核TPH2的表达,降低MAO活力达到改善抑郁症的效果。Objective To investigate the effects of moxibustion on the expression of uTptophan hydroxy- lase 2 ( TPH2 ) mRNA in median raphe nuclei and the activity of monoamine oxidase (MAO) in serum and cortex iu depression model rats and to explore its mechanism. Methods Female SD rats were randomly divided into con- trol group ( n = 6 ), model group ( n = 7 ) and moxibustion group ( n = 7 ) . The rat depression model was induced by giving isolation-housing in combination with chronic unexpected mild stress(CUMS) for 21 days. The hemi-quanti- tative reverse transcription-polymerase chain reaction(RT-PCR) was employed to detect the relative expression of TPH2 mRNA,eolorimetry was used to test the activity of MAO. Results The expression of TPH2 mRNA in medi- an raphe nuclei in rats of the moxibustion group was higher than that in the model group(relative deutisty value: (0. 3759 ± 0. 0272 ) vs (0. 2573 ± 0. 0581 ), P 〈 0.05 ). The activity of MAO in serum and cortex was reduced in rats of the moxibustion group ( ( 1.919 ± 1. 243 ) U/ml, ( 1292. 048 ± 90. 072 ) U/rag protein, respectively) compared with that in the model group( (4.117± 2. 727 )U/ml, (1441. 345±117. 050 )U/rag protein, respectively). Conclusion Moxibustion has the effects of improving depression, increasing the expression of TPH2 mRNA in median raphe nuclei and reducing the activity of MAO in serum and cortex might be one of its mechanisms.
