摘 要:目的探讨人参皂苷Rb1在大鼠脑创伤后的脑水含量及血清S100β的影响。方法参照Feeney MD等的方法建立自由落体颅脑外伤模型。将24只SD雄性大鼠随机分为假手术组、模型组、人参皂苷Rb1低剂量组、人参皂苷Rb1高剂量组,每组6只。假手术组只做开骨窗处理;人参皂苷Rb1低、高剂量组于脑创伤后即刻腹腔注射相应剂量的人参皂苷Rb1注射液;模型组在创伤后即刻腹腔注射等量生理盐水。采用干湿比重法测脑水含量,酶联免疫吸附ELISA法检测血清中S100β的质量浓度变化。结果与假手术组比较,其余3组脑含水量和血清S100β增加,有显著差异;与模型组比较,人参皂苷Rb1低、高剂量组脑含水量和血清S100β降低,有显著差异;人参皂苷Rb1高剂量组血清S100β较低剂量组显著降低。结论腹腔注射人参皂苷Rb1可减少脑创伤24 h后大鼠的脑含水量、降低血清S100β含量,提示注射人参皂苷Rb1对脑创伤后大鼠具有脑保护作用,且与剂量有关。Objective:To investigate the effect of Ginsenoside Rbl on post-cerebral-wounded rats. Methods: The Free-fall FBI model of traumatic brain injury was established according to the methods of Feeney MD and et al. 24 male SD rats were randomly divided into the shamoperated group,the model group,the low dose of Ginsenoside Rbl treatment group and the large dose of ginsenoside group. A small area of rats skull was removed only in sham-operated group. After establishing the FBI model, the same volum of saline was injected immediately into peritoneal cavity of model group rats. Different dose of Ginsenoside Rbl parenteral solution was injected im- mediately into peritoneal cavity of Ginsenoside Rbl treatment group rats. Samples were acquired after 24h of the brain injury. Brain water content was measured with the method of weighing,ELISA was used to assay the serum S100β level. Results:Brain water content and serum S100β level of rats in the model group,and 2 Ginsenoside groups were significant increased compared with the sham-operated group. Brain water content and serum S100β level of rats in 2 Ginsenoside groups were significant decreased compared with the model group. Serum S100β level of rat in the and the large dose of Ginsenoside group was significant decreased compared with the low dose of Ginseoside group. Conclusion:Ginsenoside Rbl could protect rats brain from traumatic injury by decreasing the brain water content and serum S100β level. The protective effect is related to the dose of Ginsenoside Rbl.
