摘 要:目的探讨温阳强心方对兔慢性充血性心力衰竭模型心脏结构、功能以及细胞凋亡的影响。方法将50只兔随机抽取10只为正常组,其余40只予以阿霉素静脉注射建立心力衰竭模型。造模结束后行心脏彩超检查,根据心功能分层再随机分为4组:慢性充血性心力衰竭(CHF)模型组、卡托普利组、轻剂温阳强心方组及重剂温阳强心方组。各组分别予以相应药物灌胃治疗,正常组和CHF模型组以等量的蒸馏水灌胃,每天1次,连续4周。实验结束时比较各组心脏结构和功能以及心肌细胞凋亡阳性面密度比值的变化。结果与CHF模型组比较,温阳强心方能升高心衰家兔LVEF、LVFS、E/A(P<0.01),降低IVS、LVPW(P<0.01),降低心肌细胞凋亡阳性面密度比值,差异有统计学意义(P<0.05),且重剂量组疗效优于轻剂量组。结论温阳强心方能改善阿霉素所致兔慢性充血性心力衰竭模型的心功能,抑制细胞凋亡和心室重构,且重剂量组疗效优于轻剂量组。Objective To investigate the effects of Wenyang Qiagxin decoction(WQD) on chronic congestive heart failure apoptosis and cardiac structure in rabbits.Methods Ten rabbits were randomly selected as healthy control group(HCG) and tother 40 were induced by adriamycin to prepare heart failure models.Then,according to the levels of heart functions evaluated by echocardiography,the 40 model rabbits were assigned into 4 sub-groups: modeling group(MG),captopril group(CG),WQD low dose group(LDG) and high dose group(HDG).All the animals in each treating group were given corresponding medicine respectively,and those in HCG and the MG were given the same value of distilled water administrated by gavage,once a day and lasted for 4 weeks.By the end of experimental period,all these animals were put into death to measure cardiac structure and function and to take tissue samples for determine apoptotic activity in cardiac muscle by surface density ratio of positive apoptotic cells for comparative analysis among different groups.Results Compared with that of MG,WQD significantly elevated the indicators LVEF,LVFS and E/A were(P0.01),and reduced obviously IVS and LVPW(P0.01),and in LDG and HDG decreased markedly the surface density ratio of positive apoptotic cells(P0.05),in which the effects were much more superior in HDG to that in LDG.Conclusion WQD can effectively improve the cardiac function of CCHF induced by adriamycin and significantly inhibit apoptotic activity in cardiac muscle and remodeling of cardiac ventricle among in rabbits and the effects in HDG isbetter than that in LDG.
