摘 要:目的建立用微流控芯片毛细管电泳激光诱导荧光检测盐酸阿霉素注射剂中盐酸阿霉素含量的方法。方法以荧光素钠为内标物,20 mmol/L的Na2B4O7-NaOH缓冲溶液(pH 10.0)作为电泳缓冲液,采用夹流进样和分离方式,进样电压500 v,分离电压1 800 V。结果 1.5 min内即可完成进样和分离,盐酸阿霉素质量浓度在0.58~5.8μg/mL范围内线性关系良好(r=0.996),检出限为0.17μg/mL(S/N=3),加标回收率为97.7%~101.5%,结论微流控芯片毛细管电泳法简单、快速、准确,样品用量少,可用于测定盐酸阿霉素注射剂中盐酸阿霉素的含量。Objective To establish a new method for the determination of doxorubicin in doxorubicin hydrochloride injection by microfluidic chip with laser-induced fluorescence detection.Methods Fluorescein sodium was used as the internal standard.The optimal separation conditions,such as the pH and concentration of buffer solution,separation voltage and injection time,were investigated.20 mmol/L sodium borate-sodium hydroxide was used as electrophoresis buffer solution(pH 10.0).The introduction voltage was 500 V,and the separation voltage was 1 800 V.Results The separation was finished in 1.5 min.Good linearity can be observed in a range of 0.58-5.8 μg/mL(r=0.996),and the detection limit(S/N=3) was 0.17 μg/mL and the recovery ratio was within 97.7%-101.5%.Conclusion The method established on microchip was simple,rapid,accurate,and low cost,which can be used in the assay of doxorubicin hydrochloride.
