摘 要:目的:探讨茵栀黄注射液对新生大鼠胆红素脑损伤的保护作用。方法:175只7日龄SD大鼠随机分成正常对照组、脑损伤组、治疗组Ⅰ、Ⅱ、Ⅲ,腹腔注射质量分数为120 mg/kg的胆红素溶液诱发新生大鼠脑损伤,治疗组Ⅰ、Ⅱ和Ⅲ新生大鼠建模同时每天分别腹腔注射质量分数分别为513.2、265.5和132.8mg/kg的茵栀黄注射液,持续给药2周。在给药治疗48h和2周后,重氮反应法检测脑组织中胆红素浓度,定磷法检测脑组织Na+-K+-ATP酶的活性,并在给药治疗2周后苏木精-伊红(HE)染色观察大脑皮层组织形态学的改变。结果:治疗组大鼠脑组织胆红素浓度均明显低于脑损伤组,而脑组织Na+-K+-ATP酶活性明显高于脑损伤组,且有显著的量效关系;治疗组大鼠大脑皮层组织形态结构的异常改变较脑损伤组大鼠有不同程度的改善。结论:茵栀黄注射液能显著改善胆红素脑损伤新生大鼠脑组织Na+-K+-ATP酶活性和形态结构的异常改变。Aim:To observe the protective effects of Yinzhihuang injection on bilirubin-induced braininjury in neonatal rat. Methods: 175 seven-day old neonatal rats had been randomly divided into normalcontrol group, brain injury group, therapy group Ⅰ , Ⅱ .and Ⅲ. Bilirubin solutions of 120 mg/kg massfraction were injected into the abdominal cavity of seven - day old rats for inducing brain injury. Mean-while, Yinzhihuang injection solutions of 513.2,265.5 and 132. 8 mg/kg mass fraction were injected in-to the abdominal cavity of the neonatal rats each day for treatment of the brain injury during two weeks intherapy group Ⅰ , therapy group Ⅱ and therapy group Ⅲ respectively. The bilirubin concentrations of thebrain tissues were examined via a diazo method and the Na^+-K^+-ATPase activities of the brain tissueswere analyzed by rooting phosphorus after 48 hours and 2 weeks treatment respectively. The morphologi-cal changes of cerebral cortex were observed with Hematoxylin-Eosin (HE) staining after 2 weeks treat-ment. Results: The increased bilirubin concentrations and the decreased Na^+-K^+-ATPase activities inthe brain tissues had been corrected significantly, which was characterized by significant dosage-effect re-lationship, and the abnormal morphological changes of cerebral cortex were improved at different degreein rats of the therapy groups. Conclusion:Yinzhihuang injection can significantly improve the abnormalchanges of the Na^+-K^+-ATPase activities and morphological structures of the brain tissues in neonatalrats with bilirubin -induced brain injury.
