摘  要:目的:探讨促性腺激素预处理的小鼠在卵泡发育及排卵过程中Prdx2在卵巢中的表达,为进一步研究Prdx2在调控卵泡生长、发育及成熟中的作用奠定基础。方法:将22~24d大小的75只C57/BL6雌性小鼠随机分成14组(生理盐水组10只,其余各组5只),分别为注射孕马血清促性腺激素(PMSG)后2、4、6、8、10、12、24、48h组和注射人绒毛膜促性腺激素(hCG)后5、8、10、12、24h组以及生理盐水组,其中生理盐水组分别于注射生理盐水后2h(C0,5只)及48h(C1,5只)取出卵巢。采用实时定量PCR及免疫印迹法检测Prdx2mRNA及蛋白在促性腺激素处理后不同时间点的动态表达。结果:小鼠腹腔注射PMSG后Prdx2表达均低于生理盐水组,于注射后2h表达量最低,随后表达逐渐增高,24h到达最高点,但表达仍低于生理盐水组。生理盐水组注射生理盐水后2h(C0)与注射PMSG后2、4、6、8、10、12、48h相比差异均具有统计学意义(P<0.05),而与注射PMSG后24h相比则差异无统计学意义(P=0.238)。注射PMSG后不同时间点两两比较结果表明2hPrdx2的表达同6h相比差异有统计学意义(P=0.00),其他时间点两两比较差异无统计学意义(P>0.05)。小鼠腹腔注射PMSG后48h(P48)注射hCG促排卵,促排卵后不同时间点卵巢标本mRNA表达量同P48相比差异均无统计学意义(P>0.05)。同生理盐水组注射生理盐水后48h(C1)相比差异均有统计学意义(P<0.05)。Prdx2蛋白在小鼠腹腔注射PMSG后10h表达水平最低,其他各组间差异无统计学意义。注射hCG后各时间点同生理盐水组相比,表达差异均无统计学意义。结论:Prdx2可能在小鼠卵泡发育及排卵过程中发挥重要作用。Objective To detect the ovarian expression of Prdx2 during follicle development, maturation and ovulation in the gonadotropin primed immature mice. Methods 75 immature C57/BL6 mice were randomly allocated into 14 groups (control group: 10 mice, treated groups: 5 mice/group). The expression of Prdx2 was detected by real-time PCR and Western blot at the time points of 2 h, 4 h, 6 h, 8 h, 10 h, 12 h, 24 h and 48 h after PMSG treatment, 5 h, 8 h, 10 h, 12 h and 24 h after injecting of hCG for inducing ovulation. Results The real-time PCR data showed that the mRNA level in the treated groups were significantly reduced than those in the control group after the injection of PMSG. At 2 h, its expression appeared the lowest, then increased gradually and reached a peak at 24 h, but still lower than the control group. Significant differences were found between the control group (C0, the group treated with sodium chloride for 2 h) and the groups treated with PMSG for 2 h, 4 h, 6 h, 8 h, 10 h, 12 h, 48 h, respectively(P 0.05), yet there was no significant difference between the C0 group and the 24 h group (P = 0.238). At the same time, we found that there was significant difference between the 2 h group and the 6 h group (P = 0.00), but the other groups had no significant difference compared to each other(P 0.05). The mice ovulation were induced by injecting hCG after dealing with PMSG, the expression of Prdx2 in the groups after injecting hCG were significantly downregulated when compared to the control group (C1, the group treated with sodium chloride for 48 h, P 0.05), yet had no clear difference as compared with the group of P48 (the group with PMSG for 48 h, P 0.05). Conclusion Prdx2 probably plays an important role in follicle development, maturation and ovulation.
