摘 要:目的:探讨巨噬细胞刺激蛋白(MSP)对COPD大鼠肺泡巨噬细胞氧化应激和细胞因子产生的影响。方法;培养正常和COPD大鼠肺泡巨噬细胞,予不同浓度MSP干预,采用酶联免疫法检测细胞上清液中细胞因子TNF-α、IL-8、IL-1β和IL-10的浓度,比色法检测细胞上清液中丙二醛(MDA)和超氧化物歧化酶(SOD)的水平。结果:经MSP处理后,正常大鼠和COPD大鼠肺泡巨噬细胞分泌TNF-α、IL-8、IL-1β和IL-10呈浓度依赖性增加,而COPD大鼠肺泡巨噬细胞上清液中TNF-α、IL-8和IL-1β浓度较正常大鼠增加更显著。MSP呈浓度依赖性刺激正常和COPD大鼠肺泡巨噬细胞分泌MDA增加,抑制其SOD的产生,而COPD大鼠肺泡巨噬细胞上清液中SOD水平较正常大鼠降低更加明显。结论:MSP能显著增加COPD大鼠肺泡巨噬细胞分泌TNF-α、IL-8和IL-1β,抑制其产生SOD。提示MSP在COPD的发病机制中可能起重要作用。Objective To investigate the effect of mcrophage-stimulating protein(MSP)on the release of cytokine and oxidative stress of alveolar macrophage(sAms)in COPD rats.Methods Cultured normal and COPD rats Ams were treated with different concentrations of MSP.The levels of TNF-α、IL-8、IL-1β and IL-10 in the supernatant of AMs were determined with ELISA.The levels of maleic dialdehyd(MDA)and superoxide dismutase(SOD) in the supernatant of AM were assayed by chromatometry method.Results MSP increased TNF-α、IL-8、IL-1β and IL-10 levels in the supernatant of AMs from normal and COPD rats in a concentration-dependent manner.The levels of TNF-α、IL-8 and IL1β were significantly increased in copd rats Ams,as compared to normal rats.MSP increased the production of MDA and inhibited the production of SOD in a concentration-dependent manner.The level of SOD was significantly decreased in copd rat Ams,as compared to normal rats.Conclusion MSP increases significantly the release of TNF-α、IL-8、IL-1β and inhibits the production of SOD in cultured COPD rats AMs.It is indicated that MSP plays important role in the pathogenesis of COPD.
