摘  要:目的:探讨大鼠体内雌激素对子宫动脉和冠状动脉的作用与两者的平滑肌细胞(SMC)表现型的关系。方法:HE染色观察子宫动脉和冠状动脉的的组织形态学结构。间接免疫荧光染色比较子宫动脉和冠状动脉SMC中平滑肌特异性肌动蛋白α-SM actin和非肌细胞型肌动蛋白β-NM actin平均荧光强度。Western blot检测子宫动脉和冠状动脉SMC的α-SM-actin和β-NM-actin含量,并检测β-雌二醇刺激下两者β-NM actin含量变化情况。结果:冠状动脉SMC以收缩型SMC为主,高表达α-SM actin;子宫动脉SMC以合成型SMC为主,高表达β-NMactin。在生理浓度β-雌二醇刺激下子宫动脉SMC的β-NM actin含量显著上升。结论:冠状动脉和子宫动脉的SMC表现型存在明显差异,前者显示收缩型SMC特点,后者接近合成型SMC。雌激素能够刺激合成型SMC的增殖。Objective: To explore whether the different effects of estrogen on the uterine artery and coronary artery are due to a phenotypic difference of smooth muscle cells(SMCs).Methods: Histo-morphological structure of uterine artery and coronary artery were observed by HE staining.Mean fluorescence intensity of α-SM actin and β-NM actin in the uterine artery and coronary artery SMCs were measured and compared by indirect immunofluorescence staining.Western blot was adopted to detect the content of α-SM-actin and β-NM-actin in uterine artery and coronary artery SMC,and the changes of β-NM actin after β-estradiol stimulation.Results: Most SMCs of coronary artery were contractile phenotype SMCs,with high expression of α-SM actin.Mainly SMCs of uterine artery were synthetic phenotype SMCs,with high expression of β-NM actin.The content of β-NM actin in uterine artery increased significantly after stimulated by β-estradiol.Conclusion: The SMC phenotypes are significantly different between coronary artery and uterine artery in rats.Coronary artery SMCs had characters of contractile phenotype,and uterine artery SMCs was close to the synthetic SMCs.Estrogen can stimulate the proliferation of synthetic SMCs.
