摘  要:目的评价孕早期七氟醚麻醉对子代大鼠认知功能的影响。方法孕5~7d的sD大鼠30只,采用随机数字表法,将其随机分为3组(n=10):正常对照组(C组)、吸入七氟醚4h组(s,组)和吸入七氟醚8h组(s:组)。s1组和s2组分别吸入95%氧气-1.8%七氟醚4h和8h,氧流量2L/min,C组吸入95%氧气8h。分别于出生后20、30d取5只母鼠的子鼠,采用Morris水迷宫实验测定认知功能,持续7d,然后处死子代大鼠,分离海马组织,测定N-甲基-D-天冬氨酸受体2B亚基(NR2B)mRNA及其蛋白和hippyragranin mRNA的表达水平。结果3组子代大鼠水迷宫实验结果、NR2BmR.NA及其蛋白和hippyragranin mRNA表达水平差异均无统计学意义(P〉0.05)。结论孕早期七氟醚麻醉对子代大鼠认知功能无影响。Objective To investigate the effect of sevoflurane anesthesia during the early pregnancy on the cognitive function in offspring rats. Methods Thirty SD rats at 5-7 day gestation were randomly divided into 3 groups ( n = 10 each) : control group (group C), 4 h inhalation of sevoflurane group (group SI ), and 8 h inhala- tion of sevoflurane group (group S2 ). Groups S1 and S2 inhaled 1.8% sevoflurane (in 02 2 L/min) for 4 and 8 h respectively, while group C inhaled oxygen 2 L/rain only for 8 h. At 20 and 30 days after birth, offspring rats from 5 pregnant rats were tested for learning and memory abilities using Morris water maze. After 7 days of the test, the offspring rats were sacrificed and hippocampi were isolated for determination of the expression of N-methyl-D-aspar-tate receptor subunit 2B (NR2B) mRNA and protein and hippyragranin mRNA. Results There were no significant differences in the results of Morris water maze test and expression of NR2B mRNA and protein and hippyragranin mRNA among the three groups ( P 〉 0.05). Conclusion Sevoflurane anesthesia during early pregnancy has no effect on the cognitive function in offspring rats.
