摘 要:目的探讨鞘内注射右美托咪啶对小鼠急性炎性痛的影响。方法雄性成年昆明小鼠110只,体重18~20g,采用随机数字表法,将其随机分为5组(n=22):二甲亚砜组(DMSO组)、炎性痛组(IP组)和不同剂量右美托咪啶组(D1-3组)。D1-3组分别鞘内注射右美托咪啶0.100、0.020和0.004μg/μl(溶于10μl人工脑脊液中)。于鞘内注射后5min时,IP组和D1-3组左后爪底皮内注射辣椒素0.1μg/μl(溶于10μl二甲基亚砜中),制备小鼠急性炎性痛模型;DMSO组爪底注射二甲基亚砜10μl。每组随机取8只小鼠,于爪底给药前1h(T0)、给药后即刻(T1)、5min(T2)和10min(T3)时测定热缩爪潜伏期;每组随机取8只小鼠,记录5min内出现舔/咬给药侧爪的时间。于爪底给药后5min时,每组随机处死6只小鼠,取脊髓L4-5段,采用Western blot法测定磷酸化ERK1/2(p-ERK1/2)和ERK1/2的表达。结果与DMSO组比较,IP组和D3组T1和T2时热缩爪潜伏期缩短,舔/咬爪时间延长,脊髓p-ERKl/2表达上调(P〈0.01),D1组和D1组T1-3,时热缩爪潜伏期延长(P〈0.01),舔/咬爪时间和脊髓p-ERK1/2表达差异无统计学意义(P〉0.05);与IP组和D3组比较,D2组和D2组D2组T1-3,时热缩爪潜伏期延长,舔/咬爪时间缩短,脊髓p-ERK1/2表达下调(P〈0.05);IP组和D,组间、D3组和D1组间热缩爪潜伏期、舔/咬爪时间和脊髓p-ERK1/2表达比较差异无统计学意义,各组间脊髓ERK1/2表达比较差异无统计学意义(P〉0.05)。结论鞘内注射右美托咪啶可减轻小鼠急性炎性痛,其机制与抑制脊髓ERK1/2的活性有关。Objective To investigate the effect of intrathecal dexmedetomidine on acute inflammatory pain in mice. Methods One hundred and ten aduh male Kunming mice, weighing 18-20 g, were randomly divided into 5 groups( n = 22 each) : dimethyl sulfoxide group(group DMSO) ; inflammatory pain group(group IP) and different doses of dexmedetomdine group (group D1-3 ). Intrathecal dexmedetomdine 0. 100, 0.020 and 0.004 μg/μl (in 10 fd artificial cerebrospinal fluid) were injected in groups Dl 3 respectively. Five minutes later, inflammatory pain was induced by capsaicin 0.1 μg/μl (in 10 kd DMSO) given intracutaneously in the left hindpaw in groups IP and D1-3. While DMSO 10 μl was given intracutaneously in the left himlpaw in group DMSO. Thermal withdrawal latency was determined at 1 h before (TO ), immediately, at 5 and 10 rain after intracutaneous injection (T,_3) in 8 mice in each group. Licking/biting time was recorded within 5 rain after intraeutaneous injection in 8 mice in each group. Six mice were sacrificed and L4_5 spinal cord was removed for determination of phosphorylated extracellular signal-regulated protein kinase 1/2 (p-ERK1/2) and ERK1/2 at 5 min after intracutaneous injection. Results Compared with group DMSO, the paw withdrawal latency was significantly shortened at T1 and T2, the licking/biting paw time prolonged and the expression of p-ERK1/2 up-regulated in groups IP and D3 , the paw withdrawal latency was significantly prolonged at T1-3 in groups D1 and D2 (P 〈 0.01 ) . Compared with groups IP and D3 , the paw withdrwal latency was significantly prolonged at T1-3, the licking/biting paw time shortened and the expression of p-ERK1/2 down-regulated in groups Dl and D2 ( P 〈 0.01 ). There was no significant difference in the paw with-drawal latency, licking/biting paw time and expression of p-ERKI/2 between groups IP and D3 and between groups D1 and D2 ( P 〉 0.05). There was no significant difference in the expression Of ERK1/2 among all the groups
