摘 要:目的 探讨高血压病患者血管平滑肌细胞 (VSMC)一氧化氮合酶 (NOS)活性和NOS基因表达的特点及其与高血压发病的关系。方法 采用复合胶原酶法分离培养高血压病患者和血压正常者动脉VSMC ,并以血红蛋白分光光度法、原位反转录聚合酶链反应检测VSMC的NOS活性和诱生型NOS(iNOS)mRNA表达量。结果 (1)高血压组VSMC的NOS活性和iNOSmRNA表达量均显著低于正常组 (P <0 0 1) ;(2 )高血压组和对照组VSMC的NOS活性均随培养时间的延长而显著地增强(P <0 0 1) ;(3)高血压组和对照组VSMC的NOS活性均随iNOSmRNA表达的增加而增强 ,并均呈显著的正相关 (r=0 838;r=0 975 ,P值均 <0 0 1)。结论 高血压病患者VSMC的iNOS活性下降可能是iNOS基因表达下调所致 ,这可能是高血压发病的重要病理基础。Objective To studies the relationship between the nitric oxide synthase (NOS) activity and inducible NOS (iNOS) gene expression in the arterial smooth muscle cells (VSMCs) of hypertensives and the pathogenetic role in essential hypertension (EH) Methods By separating and culturing VSMCs of EH patients and normotension (NT) subjects in vitro with multiple collagens method, NOS activity and inducible nitric oxide synthase mRNA (iNOS mRNA) expression were measured by means of reverse transcription PCR in situ and hemoglobin spectrophotometer Results (1) NOS activity and iNOS mRNA expression in VSMCs of the EH group were much lower than those of the NT group; (2) NOS activity in both the EH group and NT group increased with prolongation of culture time; (3) NOS activity in VSMCs of the EH group and NT group was enhanced with the increase of iNOS mRNA expression and there was obvious positive correlation between them ( r= 0 838, P <0 01; r= 0 975, P <0 01) Conclusion The decrease of iNOS activity in VSMCs of hypertensives may be due to depression of iNOS gene expression and this may be an important pathogenetic factor of EH
