艾灸穴位对大鼠心肌缺血-再灌注损伤心肌细胞凋亡基因的影响

摘　　要：目的:研究艾炷灸膻中、膈俞穴益气活血法预处理对大鼠心肌缺血-再灌注损伤心肌细胞凋亡基因的影响。为临床防治心肌缺血-再灌注损伤提供新的思路和方法。方法:先结扎大鼠冠状动脉左前降支(LAD),形成心肌缺血,观察线扎紧部位心肌的颜色,再灌注后取心脏进行病理学观察,免疫组化法测caspase-3、bcl-2蛋白表达。结果:(1)凋亡标志因子caspase-3在心肌缺血再灌注损伤后表达增多,艾炷灸膻中、膈俞穴益气活血法预处理可降低其表达。(2)抑凋亡基因bcl-2在心肌缺血再灌注损伤后表达减少,艾炷灸膻中、膈俞穴益气活血法预处理可使bcl-2表达增多。结论:艾炷灸膻中、膈俞穴益气活血法预处理对大鼠心肌缺血再灌注损伤起抑制作用。其作用可能通过降低caspase-3表达,增高bcl-2表达来表现。To study the effects of moxibustion pretreatment in rats with myocardial apoptosis induced by ischemia-reperfusion. Methods：The left anterior descend coronary artery was legated to establish the ischemiareperfusion heart model in rats. Immunohistochemical staining was used to detect the changes of apoptosis and protein expression of caspase-3.bcl-2. Histopathological changes in myocardium was also observed. Results： 1. The expression of caspase-3 gene increased by ischemiareperfusion and decreased by pretreatment of moxibustion of the Danzhong and Geshu points. 2. The expression of bcl-2 gene decreased by ischemia reperfusion and increased by pretreatment of moxibustion the Danzhong and Geshu points. Conclusions： Moxibustion pretreatment by nourishing the Qi and activating blood flow shows obviously preventive and therapeutic effects on reperfusion injury and could inhibit cardiomyocyte apoptosis by decreasing bcl-2 gene expression and increasing caspase-3 gene expressing.
