摘  要:目的建立高效液相色谱法测定葛根解酒片中葛根素和2,3,5,4'-四羟基二苯乙烯-2-O-β-D-葡萄糖苷(以下简称二苯乙烯苷)的含量。方法采用kromasil C18柱(250 mm×4.6 mm,5μm),流动相:乙腈(A)-水(B),梯度洗脱〔0 min,φ(B)=90%、40 min,φ(B)=85%、60 min,φ(B)=75%〕,流速:1.0 mL.min-1,检测波长:250 nm(0 min)、320 nm(30 min)。结果葛根素和二苯乙烯苷进样量分别为50.7~676.0 mg.L-1(r=0.999 6)和13.8~184.0 mg.L-1(r=0.999 5)线性关系良好,平均回收率分别为96.7%(RSD=1.7%,n=6)和96.4%(RSD=1.4%,n=6)。结论该方法可同时测定葛根解酒片中葛根素和二苯乙烯苷的含量。Objective To develop an HPLC method for determination of puerarin and 2,3,5,4'-tetrahydrox-ystilbene-2-O-β-D-glucoside in Gegenjiejiu tablets( traditional Chinese medicines). Methods Analyses were carded out using a kromasil C18 column(250 mm ×4. 6 mm,5μm). The mobile phase was A( acetonitrile)- B (water) with gradient elution [ 0min, φ ( B ) = 90 % ; 40 min, φ ( B ) = 85 % ; 60 min, φ ( B ) = 75 % ] and the flow rate was 1.0 mL.min-1 ,the UV detector was set at 250 nm(0 min) and 320 nm(30 min). Results A linear relationships of puerarin and 2,3,5,4'-tetrahydroxystilbene-2-O-β-D-glucoside were good at the range of 50. 7- 676. 0 mg. L - 1 ( r = 0. 999 6 ) and 13.8-184. 0 mg. L - 1 ( r = 0. 999 5 ), respectively. The average recovery were 96.7 % (RSD = 1.7 %, n = 6) and 96. 4 % (RSD = 1.4 %, n = 6), respectively. Conclusions The method is suitable for the simultaneous determination of puerarin and 2,3,5,4'-tetrahydroxystilbene-2-O-β- D-glucoside in Gegenjiejiu tablets.
