摘  要:目的研究葛根素对黑火药烟雾诱导支气管上皮细胞表达IL-8及IL-6基因的抑制作用。方法用黑火药烟雾刺激支气管上皮细胞,用实时PCR方法定量检测黑火药烟雾刺激后支气管上皮细胞中IL-8及IL-6基因表达水平的变化;黑火药烟雾刺激前支气管上皮细胞培养液中加入葛根素,观察葛根素对黑火药烟雾刺激支气管上皮细胞表达IL-8及IL-6基因的抑制作用。结果烟雾刺激24 h后,IL-6及IL-8基因表达分别增加1.534±0.653倍和2.231±0.054倍;支气管上皮细胞在烟雾刺激前8 h加入不同浓度葛根素培养,黑火药烟雾诱导IL-6和IL-8的基因表达被显著抑制,并呈剂量依赖性。结论黑火药烟雾可诱导支气管上皮细胞炎性细胞因子IL-6、IL-8 mRNA表达增加,葛根素可抑制黑火药烟雾刺激支气管上皮细胞炎性细胞因子IL-6、IL-8 mRNA表达。Objective To study the inhibition effect of puerarin on gene expression of IL-8 and IL-6 in BEAS-2B cells stimulated by black powder smoke.Methods BEAS-2B cells were stimulated by black powder smoke.The gene expression of IL-8 and IL-6 of the cells was determined by real-time PCR.Puerarin was added to BEAS-2B cells before being stimulated by black powder smoke.The gene expression of IL-8 and IL-6 in BEAS-2B cells injured by black powder smoke was detected.Results After being injured by black powder smoke for 8 h,the gene expression of IL-8 and IL-6 of BEAS-2B cells was up-regulated(1.534±0.653 fold and 2.231±0.054 fold respectively).When different concentrations of puerarin were added to BEAS-2B cells 24 h before smoke stimulation,the gene expression of IL-6 and IL-8 in the cells was significantly inhibited in a dose-dependent manner.Conclusion Black powder smoke increases the gene expression of inflammatory cytokine IL-6 and IL-8 and this is inhibited by puerarin effect.
