替考拉宁分离条件的响应面法优化

摘  要:利用大孔吸附树脂分离纯化替考拉宁,并对其分离条件进行优化。以替考拉宁发酵液提取粗品为原料,研究上样浓度、解吸剂、解吸剂pH、解吸剂浓度等因素对替考拉宁含量的影响。在单因素试验基础上对上样浓度、解吸剂pH、解吸剂浓度三个因素进行Box-Behnken试验设计,以替考拉宁含量为响应值,利用响应面分析法对分离条件进行优化。在上样浓度为2500μg/mL、解吸剂pH2.0、解吸剂浓度40%的分离条件下,替考拉宁含量达90.8%。响应面法优化替考拉宁含量比优化前的单因素试验结果有较大提高。Studies were made on separated crude teicoplanin by macroporous resin and optimized the separated factors, and the effect of different sample concentration, desorption agent, desorption agent pH, desorption agent concentration on yield of the teicoplanin content. The Box-Behnken design was applied to fit a response surface function, which based on the one variable-at-time design. The optimum separating conditions were got through the function. The optimal levels for obtaining 90.8% was as follows: the sample concentration is 2500μg/mL, desorption agent pH is 2.0, desorption agent concentration is 40%. The Box-Behnken design can be used on the separating teicoplanin,and the result is better than that in the one-variable-at-time design.
