摘 要:对凡纳滨对虾(Litopenaeus vannamei)血细胞进行离体镉离子(Cd2+)应激试验。Cd2+浓度为10-3~10-9M,在应激6h后取样,利用流式细胞术(FCM)检测血细胞的活性和活性氧(ROS)含量。结果显示,当Cd2+浓度为10-5M时,血细胞的ROS含量显著升高;当Cd2+浓度为10-3和10-4M时,血细胞的死亡率和ROS含量均显著上升;表明Cd2+的细胞毒性作用与其浓度相关,Cd2+浓度达到10-5M时即可诱导血细胞ROS的产生,但当Cd2+浓度达到10-4M及以上时,被诱导产生的ROS对细胞造成氧化伤害,从而导致细胞死亡。Stress test of cadmium ion on haemocytes of Litopenaeus vannamei in vitro was conducted. Exposed to concentration of 10^-9 - 10^-3 M Cd^2+ in 6 h in vitro, haemocyte viability and reactive oxygen species (ROS) of L. vannamei haemocyte samples were determined through flow cytometry (FCM). The results showed that ROS con- tents in haemocytes surged significantly under the concentration of 10^-5 M cadmium; Under the concentration be- tween 10^-4 and 10^-3 M cadmium, mortality and ROS contents of haemocytes ascended emphatically respectively. These results showed the cytotoxicity of Cd^2+ on L. vannamei was in relevance to the concentration of Cd^2+. ROS of haemocytes would be induced with Cd^2+ concentration reaching 10^-5 M, but the induced ROS would cause oxida- tive damage to haemocytes when Cd^2+ concentration reaching 10^-4 M or above, thus contributing to haemocyte death.
