摘 要:目的:观察比较关木通及当归四逆汤各拆方组中马兜铃酸A的变化趋势,探讨中药配伍对马兜铃酸A含量的影响。方法:参照当归四逆汤原方比例将供试样品分6组:关木通组、全方组、关木通+补气补血组、关木通+散寒组、关木通+甘草组、阴性对照组(全方去关木通);采用反相高效液相色谱(RP-HPLC)法,以甲醇—水—冰乙酸(体积比74∶24∶1)为流动相,以DiamonsilTM(钻石)C18(250 mm×4.6mm,5μm)为色谱柱,检测波长237 nm,测定各组中马兜铃酸A的含量。结果:与关木通组相比,当归四逆汤全方中马兜铃酸A含量最低;关木通+补血组马兜铃酸A含量也有明显降低。结论:在方剂配伍理论指导下,通过合理配伍降低关木通的毒性是可行的。Objective : To compare aristolochic acid (AA) content in Caulis Aristolochiae Manshuriensis (CAM) and in the separated prescriptions of Danggui Sini Decoction (I)SD) , and to explore the effect of Chinese herbal medicine compatibility on AA content. Methods: According to the herbs percentage compared to DSD, the herbs samples were named : CAM group, DSD group, CAM group with Qi-blood - nourishing herbs, CAM group with herbs for cold scattered, CAM group with Radix Glycyrrhizae, and DSD group with CAM removed. The AA content was determined by reverse phase HPLC. Gradient eluted with a mixture of methyl alcohol water and acetic acid ( volume proportion as 74 : 24 : 1 ) , the chromatographic conditions were as follows: Cls Diamonsihm analytical column, and detection wavelength at 237nm. Results : AA content was the lowest in DSD group, and was lower in CAM group with yin - nourishing herbs than that in CAM group. Conclusion:Subjecting to the theory of Chinese herbal medicine compatibility, it is feasible to reduce the CAM toxicity by composing a prescription with proper proportion of herbs.
