摘 要:目的:优化天仙子多糖的提取工艺,建立含量测定方法。方法:采用L9(3)4正交试验,以多糖含量为评价指标,确定多糖的提取工艺,并采用苯酚-硫酸法,测定多糖的含量。结果:最佳提取工艺为100℃,加水10倍量回流提取3次,每次1h;葡萄糖的浓度在0.1202~0.4206 mg/mL范围内呈良好的线性关系,回收率为99.79%,RSD=0.84%。结论:确定的工艺稳定可行,为天仙子多糖的进一步研究奠定基础。Objective : To optimize the extraction technology and establish the determination method of polysaccharides in Hyoscyamus niger L. Methods : Phenol - sulphate colorimetry was used to determine the content of polysaccharides. According to the yield of polysaccharides, the orthogonal design was used to optimize the extraction technology of polysac- charides. Results: The optimum extraction procedure was adding 10 folds of water, refluxing and extracting at 100% for 3 times, lh for each time. The concentration of polysaccharide was linear in the range of 0. 1202 -0. 4206 mg/mL. The average recovery was 99.79%, RSD = 0.84%. Conclusion. The optimum extraction procedure is simple and reliable, which provides a basis for the further study on polysaccharides in Hyoscyamus niger L.
