摘  要:在(25±1)℃水温条件下,分别采用口灌和浸浴的给药方式,以20 mg/kg的单剂量混饲口灌及5 mg/L的剂量浸浴18 h给予噁喹酸后,用高效液相色谱法测定血浆和肌肉中的药物浓度,研究了不同给药方式下噁喹酸在欧洲鳗鲡(Anguilla anguilla)体内的药代动力学特征。结果表明:欧洲鳗鲡口灌给药后,其血浆和肌肉中噁喹酸经时过程均符合一级吸收一室开放模型,其理论方程分别为:C血浆=8.0560(e-0.00181t-e-0.00250t),C肌肉=2.9347(e-0.00176t-e-0.00217t);浸浴给药方式下,其血浆和肌肉中噁喹酸经时过程均符合一级吸收二室开放模型,其理论方程分别为:C血浆=1.8100e-0.0042t+0.5548 e-0.00143t-2.70e-0.3007t,C肌肉=1.2779e-0.0082t+0.0394 e-0.000721t-6.35 e-0.1202t,以50μg/kg为最高残留限量计算,在本实验条件下,以口灌给药方式,建议噁喹酸在欧洲鳗鲡体内的休药期不低于114 d;以浸浴给药方式,建议其休药期不低于71 d。At water temperature of (25 + 1 ) ℃, Anguilla anguiUa was administered orally with oxolinic acid (OA) in sin- gle dose of 20 mg/kg or by medicated bath in 5 mg/L for 18 hours. The concentration of OA in the plasma and muscle of the eels were determined by high performance liquid chromatography and the pharmacokinetics of OA in eels in different ways of administration were studied. The results showed that after oral administration, the plasma and muscle concentra- tion-time course of OA conformed to a one-compartment open model with the first order absorption. The pharmacokinetic e- quations were as follows : Cplasms = 8. 0560( e -0.001811 _ e -0.00250t ), Cmuscle = 2. 9347 ( e -0.00176t - e -0.00217t ) ; after medicated bath, the plasma and muscle concentration-time course of OA both conformed to a two-compartment open model with the first order absorption. The pharmacokinetic equations were as follows, : Cplasraa= 1. 8100e-0.0042t + 0. 5548 e-0.00143t _ 2.70 e-0.3007t, Cmuscle =1.2779e--0.0082t +0.0394 e-0.000721t -6.35 e-0.1202t According to the maximum residue limit(MRL) of 50 ug/kg, the withdrawl period should be no less than 114 days under oral administration and no less than 71days under medicated bath.
