摘 要:以山茱萸的叶片,茎段,种子胚等不同部位材料作为外植体进行了山茱萸不同外植体的离体培养研究,比较了不同培养条件对山茱萸疏松愈伤组织诱导和生长效果的影响,并就继代培养过程中,不同培养基组分及培养条件对其生长的影响情况进行了试验与比较分析。结果表明:KT(3 mg/L)+2,4-D(1 mg/L)对嫩叶的诱导效果最好;在光照强度1 500~2 000 lx,3%蔗糖,pH值为5.8左右时,山茱萸细胞生物量的增长率达到最大;合适的继代周期为20~25 d。Different parts of the Comus officinalis Sieb, such as leaves, stems and embryos, were taken as the explants to conduct in vitro culture, so that the impacts of different culture conditions on induction and growth of loose callus of Comus officinalis Sieb were compared, and the impacts of different components in culture medium and different culture conditions on growth of loose callus of Comus officinalis Sieb during subculture were also compared. The results showed that KT (3 mg/L)+2,4-D (1 mg/L) received best marks on induction of tender leaves; when lighting intensity was 1 500- 2 000 ix, concentration of sucrose was 3%, and pH was around 5.8, the growth rate of cell biomass in Comus officinalis Sieb reached the maximum with appropriate subculture period of 20-25 d.
