衰老进程中机体免疫器官和免疫细胞因子变化及健脾补肾中药的干预研究

摘　　要：目的观察衰老进程中白细胞介素-2(IL-2)、白细胞介素-6(IL-6)水平和胸腺、脾脏指数的变化及健脾补肾中药干预的效应。方法将大鼠随机分为空白对照组,模型组,健脾补肾中药大、中、小治疗组,阳性对照组,每组8只。除空白对照组外,其余大鼠以D-半乳糖0.125 g/(kg.d)连续40 d皮下注射建立亚急性衰老大鼠模型,造模成功后健脾补肾中药组采用不同剂量健脾补肾中药干预,阳性对照组予桂附地黄丸干预。40 d后测定各组大鼠血清IL-2、IL-6水平和胸腺、脾脏指数。结果模型组大鼠血清IL-2和胸腺、脾脏指数显著降低(P<0.05),血清IL-6显著升高(P<0.05);健脾补肾中药组大鼠血清IL-2和胸腺、脾脏指数增高(P<0.05),血清IL-6显著降低(P<0.05)。结论健脾补肾中药具有调节衰老机体细胞因子失衡、抑制免疫器官退化和萎缩的作用。Objective To investigate the variation of IL-2 and IL-6 in serum and thymus and spleen index of aging,and effect of Chinese medicine with spleen-strengthening and kidney-tonifying on them.Methods Rats were randomly divided into control group,model group,treatment group of different dose Jianpi Bushen herbs and positive control group,8 rats in each.The subacutely aging model rats were made by subcutaneous injecting D-galactose 0.125 g/（kg？d） continually for 40 days.After treatment with different dose Jianpi Bushen herbs for 40 days,levels of IL-2 and IL-6 in serum and thymus and spleen index were evaluated.Results The level of IL-2 in serum,thymus and spleen index in model group were downregulated（P〈0.05）,level of IL-6 in serum was increased significantly（P〈0.05）.After treated with spleen-strengthening and kidney-tonifying medicine,the level of IL-2 in serum,thymus and spleen index were upregulated（P〈0.05）,level of IL-6 in serum was reduced significantly（P〈0.05）.Conclusion Chinese medicine with spleen-strengthening and kidney-tonifying can adjust and recover the unbalance of cytokines,and also has good effect on immune organs and functions in aging process.
