油酸性急性肺损伤家兔γ-干扰素的表达与膈神经放电曲线变化的关系

摘　　要：目的探讨家兔油酸性急性肺损伤时膈神经放电曲线变化与γ-干扰素(IFN-γ)表达的关系。方法健康新西兰家兔40只,随机分为复制油酸性急性肺损伤模型组(实验组)和对照组,每组20只,分别测定实验组和对照组家兔动脉血氧分压(PaO2)、外周血和支气管肺泡灌洗液(BALF)中IFN-γ表达水平及膈神经放电曲线,并进行对比分析。结果实验组PaO2低于对照组(P<0.05)。实验组BALF中和外周血中IFN-γ的表达水平高于对照组(P<0.05),在肺组织中IFN-γ的表达高于对照组(P<0.05),家兔膈肌放电曲线幅度和对照组家兔相比明显减小,放电持续时间缩短,频率减少(P<0.05),而且实验组家兔PaO2数值与膈神经放电曲线的幅度、放电持续时间、放电频率呈正相关(r=0.1826,P<0.05)。结论 IFN-γ参与油酸引起的肺部急性炎症反应,放大炎症反应,导致急性肺泡-毛细血管膜损伤,膈神经放电减弱。【Objective】 To study the relationship between expressions of IFN-γ and change of phrenic nerve discharge curve in rabbits with oleic acid induced acute lung injury.【Methods】 Forty New Zealand rabbits were random divided into experimental group（EG） and control group（CG）.In EG group,oleic acid was offered to induce acute lung injury.The level of IFN-γ in serum and bronchial alveolar lavage fluid（BALF）,arterial partial pressure of oxygen（PaO2）,and the change of phrenic nerve discharge curve were measured in all rabbits of two groups.All statistics were contrasted between EG and CG.【Results】 The statistics of PaO2 in EG were lower than CG（P 0.05）.The level of IFN-γ in EG is higher than them in CG in serum and BALF（P 0.05）.The expression of IFN-γ protein of EG was more than CG in lung tissue.The duration,amplitude and interval of phrenic nerve discharge curve were reduced in EG（P 0.05）.The statistics of PaO2 was significantly negative correlated with the change of phrenic nerve discharge curve in EG（r =0.1826,P 0.05）.【Conclusion】 IFN-γ is one of the factors that cause acute inflammatory reaction in lung,and IFN-γ may enhance the inflammatory reaction.These can cause acute alveolar-capillary membrane injury,and reduce PaO2.All the above cases can reduce phrenic nerve discharge.
