摘 要:目的:探讨体外分层共培养条件下大鼠激活态雪旺细胞对人脐带间充质干细胞(hUCMSCs)生长及分化的影响。方法:分离、培养人脐带间充质干细胞和大鼠激活态雪旺细胞,通过Millicell小室实现两者体外共培养,根据共培养与否分为共培养组与对照组,通过巢蛋白(Nestin)、神经丝蛋白200(NF200)免疫荧光染色分析干细胞分化情况。结果:共培养组两种细胞在Millicell小室分层共培养条件下,hUCMSCs逐渐出现类似神经细胞的形态,对照组形态无明显变化。共培养组中hUCMSCs表达神经干细胞标志物Nestin,共培养1d后逐渐降低,而对照组仅低表达Nestin,共培养8h^7d,两组间差异有统计学意义(P<0.01);共培养组细胞共培养1d后hUCMSCs表达神经元标志物NF200,而对照组始终不表达NF200。结论:人脐带间充质干细胞有向神经细胞分化的能力,分层培养条件下大鼠激活态雪旺细胞可促进其神经方向分化。Objective:To study the influence of activated Schwann cells?in?layered co-culture on?growth and differentiation of human umbilical cord mesenchymal stem cells (hUCMSCs). Methods:The hUCMSCs and activated Schwann cells were isolated and cultured, which were divided into co-culture group and control group. These two types of cells were co-cultured by Millicell inserts in vitro. The differentiation of hUCMSCs was analyzed using nestin and NF-200 immunofluorescence staining. Results:In Millicell inserts layered co-culture conditions,UCMSCs showed neuronal morphology gradually, but no change was found in control group. The neural stem cell marker Nestin was expressed in hUCMSCs of co-culture group, which was decreased gradually after 1d. But only low level of Nestin was expressed in control group. There was a significant difference between two groups after 8 h-7 d co-culture (P 〈 0.01). The neural marker NF200 was expressed in hUCMSCs after 1 d co-culture in co-culture group,but no any NF200 expression in control group. Conclusion:hUCMSCs was able to transdifferentiate into neural cells,which can be induced by co-culture with activated Schwann cells.
