成年猪半月板体外器官培养模型的建立

摘　　要：[目的]观察成年猪半月板在体外器官培养条件下的组织变化特点,为在体外器官培养下进行半月板研究提供实验依据。[方法]成年猪8头,取双膝内、外侧半月板,从每个半月板体部横行切取宽约8 mm标本1份,共32份标本,进行体外器官培养。分别于培养0、2、4、6周后随机取出8个标本行组织学检查,HE染色:观察半月板内侧1/3区域,计数每个视野的细胞数,比较各时间点间的差异;番红O染色:观察半月板内侧1/3区域,记录染色程度,半定量评分,比较各时间点间的差异。[结果]培养0、2、4、6周后,各时间点间每高倍视野的细胞数有显著性差异(P<0.05),两两比较均有统计学差异(P<0.05)。培养0、2、4、6周后,各时间点间番红O染色半定量评分有统计学差异(P<0.05),两两比较均有统计学差异(P<0.05)。[结论]在体外器官培养模型中进行半月板的短期研究是可行的,随培养时间延长,半月板软骨细胞密度及活性逐渐下降。[Objective]To observe the histological changes of mature porcine menisci in an organ culture system in vitro.It was hoped that the experiment can provide evidence for the study of menisci in an organ culture model in vitro.[Method]Eight mature swines were involves in the study.Thirty-two meniscal specimens with 8 mm in width were obtained from each meniscus in the posterior extremities.The specimens were incubated using an organ culture model in vitro.At 0,2,4,and 6 weeks after culture,8 specimens were observed.The cell density of the inner 1/3 zone was measured using Hematoxylin eosin staining.The degree of staining in the inner zone was measured using Safranin-O staining and semiquantitative scores were obtained.[Result]At 0,2,4 and 6 weeks,the cell density were 45.88±3.69/HPF,25.75±3.30/HPF,16.38±2.67/HPF,and 7.75±1.87/HPF.Statistically significant differences were found between different time points（P〈0.05）.Paired comparison showed statistical differences（P〈0.05）.At 0,2,4 and 6 weeks,the semiquantitative scores on Safranin-O staining were 1.86±0.35,1.38±0.52,0.88±0.35 and 0.38±0.52.Statistically differences were found between different time points（P〈0.05）.Paired comparison showed statistical differences（P〈0.05）.[Conclusion]Organ culture model in vitro is feasible for the study of menisci in a short-term observation.The cell density and vialibity diminished with the passing of cultural time.
