敌百虫对大鼠大脑皮质神经细胞的形态学损伤

摘　　要：为探讨敌百虫对大鼠大脑皮质神经细胞的毒性作用,本研究在建立体外培养新生24h内SD大鼠大脑皮质神经细胞模型的基础上,在细胞培养液中添加0,5,20,80,100,200mg/L敌百虫染毒,于染毒后不同时间观察神经细胞形态,测定胞体短直径、突起长度。结果表明:①染毒12,24h均可不同程度抑制大脑皮质神经细胞的增殖,染毒组的细胞存活率均低于对照组;②染毒12h使细胞超微结构发生改变,表现细胞核皱缩、线粒体嵴模糊、线粒体肿胀变形;③染毒12h使细胞凋亡率明显增加;Hoechst 33258荧光染色,染毒组细胞核皱缩,呈新月形甚至核碎裂等典型的凋亡特征。表明不同质量浓度的敌百虫暴露可致大鼠大脑皮质神经细胞存活率降低,细胞形态损伤,细胞凋亡率升高。To investigate the effect of low concentration of trichlorfon on rat cortical neuron,based on the established model of cortical neuron cultured in vitro which were obtained from the neonate Sprague-Dawley rats within 24 h,the cell culture medium was exposured with trichlorfon of 0,5,20,80,100,200 mg/L,and then at different times observed the morphology of cortical neuron and evaluated the changes of short diameter of the cell body,neurite length.The results were as follows：①exposured with trichlorfon for 12 h and 24 h,the proliferation of cortical neuron was inhibitted in different degrees.And the cell survival rates of exposured groups were lower than those of control groups.②exposured with trichlorfon for 12 h,the ultrastructure of cells was changed with the nuclei shrinkage,mitochondria blurred and mitochondria bloated.③neurons were treated by different concentrations of trichlorfon for 12 h,the apoptosis rate increased significantly.And when lucifugal incubated with Hoechst 33258,the typical characteristic of apoptosis was observed,such as the nucleus crimpled and crescent,even nucleus disintegrated.The results showed that exposured with different concentrations of trichlorfon can lead to rat cortical neuron′s viability decreased,morphology injuried,apoptosis rate increased.
