摘  要:目的:探讨低剂量十溴联苯醚(BDE-209)单独或联合四溴联苯醚(BDE-47)对新生鼠神经干细胞增殖形态的影响。方法:取新生24h内SD大鼠海马组织进行体外神经干细胞培养,神经干细胞用Nestin免疫荧光法鉴定。实验分组为:1‰DMSO对照组,0.6、1.0、6.0μg/mL BDE-209组,3.2μg/mL BDE-47组,0.6μg/mL BDE-209+3.2μg/mL BDE-47组,1.0μg/mL BDE-209+3.2μg/mLBDE-47组,6.0μg/mL BDE-209+3.2μg/mL BDE-47组,分别对海马神经干细胞进行体外培养染毒,72h后分别测量各组神经球直径及数目。结果:新生鼠海马组织的神经干细胞巢蛋白阳性。培养72h后,剂量0.6μg/mL BDE-209组形成神经球个数与对照组无显著差异。随着染毒剂量的增加,神经球数目变少。剂量0.6、1.0μg/mL BDE-209组形成神经球大小与对照组无显著差异。BDE-209和BDE-47联合染毒对形成神经球个数的影响存在交互作用(P<0.05)。结论:一定剂量的BDE-209和BDE-47可影响神经干细胞的增殖形态,且两者之间存在一定的交互作用。Objective To study the effects of low dose BDE-209 single exposed or combined with BDE-47 on the morphology of neural stem cells (NSCs) of neonatal rats in vitro. Methods NSCs of hippocampal tissues of 24-hour old neonatal rats were obtained and cultured in vitro, NSCs were identified by nestin immunofiuorescent staining method.The experiment groups were divided into control group (1%o DMSO), BDE-209 group at doses of 0.6 μg/mL, 1.0 μg/mL, 6.0 μg/mL respectively, BDE-47 group at doses of 3.2 μg/mL, combined group: 0.6μg/mL BDE-209 + 3.2 μg/mL, BDE-47; 1.0 μg/mL BDE-209 + 3.2 μg/mL BDE-47 and 6.0 μg/mL BDE-209 + 3.2μg/mL BDE-47, After 72 hours culture exposed to BDE-209, BDE-47 or BDE-209 combined with BDE-47, the diameter and the number of neurospheres in each group were measured. Results The Nestin protein was positive in NSCs of hippocampal tissues of 24-hour old neonatal rats. After 72 hour culture, the number of neurospheres in 0.6μg/mL BDE-209 group was not significantly different from the control group, while the number decreased as the dose increased. The diameters of neurospheres in the 0.6μg/mL, 1.0 μg/mL groups were not significantly different from the control group. The combination of BDE-209 with BDE-47 on the number of neurospheres presented to be interaction (P 〈 0.05). Conclusion BDE-209 and BDE-47 can change the morphology of NSCs at certain doses, and interaction is seen between BDE-209 and BDE-47.
