摘 要:目的观察石菖蒲两种活性成分单体β-细辛醚、丁香酚及其不同比例配伍对东莨菪碱致痴呆模型小鼠学习记忆功能的影响。方法采用Morris水迷宫筛选出学习记忆成绩及体重相近的小鼠99只,随机区组分为9组,即正常对照组、模型组、安理申阳性药对照组、β-细辛醚组、丁香酚组、β-细辛醚与丁香酚的配伍比例为1∶1,3∶1,5∶1,7∶1的配伍组,每组11只。除正常对照组和模型组给予灌服相同体积生理盐水外,其余各组均给予灌服相应的治疗药液;行为学测试前除正常对照组外,其余各组均于每次测试前30 min予腹腔注射氢溴酸东莨菪碱(3 mg.kg-1)制备学习记忆障碍痴呆模型。灌胃第8天开始通过Morris水迷宫测试小鼠的学习记忆能力,以逃避潜伏期(s)和游泳路程(cm)作为观测指标。结果与正常对照组(逃避潜伏期16.55±12.99,游泳路程250.6±199.0)相比,模型组的逃避潜伏期(38.73±35.45)和游泳路程(555.7±474.9)均明显延长(P<0.01);与模型组相比,β细辛醚组(潜伏期18.63±9.18,路程290.5±145.8)、丁香酚组(潜伏期22.78±12.01,路程351.2±182.0)、3∶1配伍组(潜伏期19.25±14.43,路程312.2±246.7)及5∶1配伍组(潜伏期18.27±15.87,路程295.7±269.8)的逃避潜伏期和游泳路程均明显缩短(P<0.01,P<0.05)。结论石菖蒲中两种主要活性成分β-细辛醚与丁香酚的较优配伍比例可能在3∶1和5∶1之间。Objective To investigate the effects of two active components of Rhizoma Aeori Tatarinowii, β-asarone and eugenol, and their compatibility at different ratios on learning and memory abilities in scopolamineinduced de mentia mice. Methods Ninetynine mice with the similar weight and learning and memory ability evaluated by Morris water maze test were randomly divided into 9 groups(n=ll): normal group, model group, Donepezil hydrochloride group, 13-asarone group, eugenol group, and compatibility groups of β-asarone and eugenol in the ratio of 1 : 1, 3 : 1, 5 : i and 7 : 1. Mice were given gastric gavage of the same volume of normal saline or drugs, respectively. On the 8th day, learning and memory impairment mice models were induced by intraperitoneal inieetion of seooolamine(3 mg· kg-1) 30 minutes before behavior test. The learning and memory abilities were evaluated by Morris water maze test, with escape latency and swimming distance as observation indexes. Results Compared with normal group, mod el group showed significantly prolonged escape latency and swimming distanee(P 〈 0.01). In 13-asarone group, eugenol group and two groups of compatibility in the ratio of 3 : 1 and 5 : 1, the swimming time and distance were signifi cantly decreased compared with those in the model group(P 〈 0.01, P 〈 0.05). Conclusion The optimal compatibili ty ratio of β-asarone and eugenol which are the main two active components of Rhizoma Acori Tatarinowii may be from3 : 1 to5 : 1.
