含石菖蒲复方煎剂中β-细辛醚的含量测定研究

摘　　要：目的通过测定治疗老年性痴呆症的含石菖蒲临床复方煎剂中β-细辛醚的含量范围,为石菖蒲的安全使用提供初步数据参考。方法从文献中选用指迷汤、还少丹、黄连解毒汤3个复方,参照临床的实际煎法进行陶瓷保健壶、圆底烧瓶、陶瓷药煲3种煎煮工具进行煎煮,采用HPLC测定复方中β-细辛醚的含量。以Agilent ZORBAX ODS为色谱柱;甲醇-水(60∶40)为流动相;检测波长为253 nm;流速为1 mL.min-1;柱温为室温。结果在日服临床用量中,指迷汤中β-细辛醚含量为33.30～57.53 mg,还少丹中β-细辛醚的含量为26.61～64.84 mg,黄连解毒汤中β-细辛醚的含量为25.18～50.51 mg。结论治疗老年性痴呆中含有石菖蒲的临床复方中β-细辛醚含量范围约为25～65 mg,远低于报道的大鼠口服β-细辛醚LD501010 mg.kg-1,为石菖蒲的安全使用提供初步数据参考。Objective To determine the content range of β-asarone in the Chinese herbal compound decoction 1 Rhizoma acori tatarinowii, which was used for the treatment of Alzheimer＇s disease. Methods Zhimi decoctionI 1 Huanshao Dan, Huanglianjiedu decoction were selected from the literature. The compounds were decocted in autoI / matic pottery heahhpot, round flask and ceramic drug pot respectively. HPLC was adopted to determine the content β-asarone in the Chinese herbal compound, Agilent ZORBAX ODS column was employed with methanol-water（6 ： 40） as a mobile phase and detection wavelength at 253 nm and flow speed at 1 mL·min-1 Results The contenl of β-asarone in Zhimi decoction, Huanshao Dan and Huanglianjiedu decoction were in the range of 33.30 57.53, 26.61-64.84 and 25.18-50.51 mg in the daily dose. Conclusion The content range of β-asarone in the Chi nese herbal compound decoction of Rhizoma acori tatarinowii was 25-65 mg, which is much lower than the reporte dosage in rat oral β-asarone LDs01010 mg＇kg-1. It can provide preliminary data reference for the safe use of Rhizotr acori tatarinowii.
