摘 要:目的研究黄连生物碱的最佳提取和纯化工艺。方法以黄连总生物碱得率为指标,采用单因素试验与正交试验优选黄连总生物碱提取工艺,并采用单因素试验优选黄连总生物碱纯化工艺。结果黄连总生物碱的最佳提取工艺为:黄连饮片加8倍量1.5%的硫酸溶液,煎煮3次,每次40min;最佳纯化工艺为:黄连提取液用Ca(OH)2调节pH5。6,抽滤,滤液加热浓缩至浓度为每mL含0.24g生药,抽滤,滤液加盐酸调节pHi.5~2,加5%氯化钠溶液使溶解,冰箱中静置过夜,抽滤,滤渣真空干燥(50℃),收集纯化物。结论所建立的黄连总生物碱提取纯化工艺简单、稳定、可行。Objective To study the optimal conditions of extraction and purification process for total alkaloid in Rhi zome Coptidis. Methods With the yield ratio of total alkaoid as the assessment index, we optimized the extraction technology of total alkaloids by single factor test and orthogonal test, and optimized the purification technology of total alkaloids by single factor experiments. Results The optimal extraction technique was as follows: refluxing extraction 3 times by 1.5 % sulfur acid with liquid-material ratio at 8 : 1, 40min every time. The optimal purification process was as follows: adjusting extraction liquid to pH value being 5~6 by Ca(OH)2, filtrating, concentrating filtrate (0.24 g medicinal herbs/mL) by heating, filtrating again, adjusting the filtrate to pH value being 1.5-2 with hy drochloric acid(HC1), adjusting the salinity of solution up to 5 % by sodium chloride, keeping the solution in the re frigerator for one night, and then filtrating, drying the residue at 50 ℃(vacuum). Conclusion The optimal extraction and purification process of total alkaloids from Rhizoma Coptidis is simple, stable and feasible.
