摘 要:[目的]研究黄芪多糖的单糖组成。[方法]以2 mol/L硫酸水解黄芪多糖,然后采用吡啶溶解干燥的水解产物,结合三甲基硅烷进行衍生,最后用毛细管气相色谱法测定黄芪多糖的单糖组成。[结果]鉴定出黄芪多糖中含有阿拉伯糖、果糖、葡萄糖和甘露糖,并测定计算出阿拉伯糖-果糖-葡萄糖-甘露糖的摩尔构成比例为1∶10.309∶24.667∶0.462;另外还鉴定出了3种未知物,他们的相对保留时间分别为10.627、14.783和18.249 min。[结论]该方法简便灵敏,可以用于黄芪多糖中单糖的组分分析。[ Objective ] To research on the composition of monosaccharide from RADIX ASTRAGALI polysaccharides. [ Method ] RADIX AS- TRAGALI polysaccharides sample was hydrolyzed with 2 mol/L sulfuric acid. Then, the dried hydrolysate was dissolved in pyridine, and derivatization was conducted by trimethylsilyl ether. Finally, the composition of monosaceharide from RADIX ASTRAGALI polysaceharides was detected by Capillary Gas Chromatography. [ Result] Arabinose ,fructose, glucose ,mannose were identified from the RADIX ASTRAGALI polysaceharides. And their molar ratio calculated according to the equation was 1:10. 309:24. 667:0. 462. Besides, three unknown matters were detected, with the retention time being 10. 627, 14. 783 and 18. 249 rain. [ Conclusion] This method was simple and sensitive, and could be used for the comr)onent analvsis of monosaccharide in RADIX ASTRAGALI oolvsaccharide.
