摘 要:为探索银翘天甘醇提物含药血清的制备工艺,采用单因素试验优化采血时间、给药次数及给药剂量,于277nm检测小鼠血清中连翘苷含量,经中心组合设计与响应面分析获得银翘天甘含药血清最佳制备工艺条件。结果表明,制备银翘天甘含药血清最佳工艺条件是以0.01g/mL银翘天甘醇提物给药4次,于最后一次给药后31min采血获得的含药血清中连翘苷含量最高,达到193.788μg/mL。通过验证试验,此模型确认有效。To explore optimization of preparation technology on medicated serum of Yinqiaotiangan in mice.Methods: Based on the single factor tests,bleeding time,administration frequency,administration dose on forsythin concentration were analyzed and forsythin concentration was detected in mice serum by ultraviolet spectrophotometer at 277 nm.Then through central-composite test design and response surface methodology obtained the preparation technology conditions on medicated serum of Yinqiaotiangan.The results showed that the preparation technology conditions were bleeding time 31 minuts,administration frequency 4 times and administration dose 0.01 g/mL.In this condition,the forsythin concentration reached peak 193.788 μg/mL in mice serum.The adequacy of this model was confirmed by means of validation experiments.
