摘  要:目的探讨肌肽(carnosine)对高糖诱导的大鼠肾小球系膜细胞(rat mesangial cells,RMCs)增殖的影响及作用机制。方法以RMCs为研究对象,分别采用MTT法和BrdU-ELISA法观察肌肽对高糖诱导的RMCs增殖的影响;乳酸脱氢酶(LDH)法检测肌肽对细胞的毒性作用;酶生化法测定丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH)含量及细胞内超氧化物歧化酶(SOD)活性等。结果肌肽可显著抑制高糖诱导的RMCs增殖,在5~30 mmol/L范围内呈一定的浓度依赖效应,且与其抗氧化作用密切相关。结论肌肽通过抗氧化作用抑制高糖诱导的RMCs增殖,且有浓度依赖趋势。Objective To study the effect of carnosine on cell proliferation in rat glomerular mesangial cells(RMCs) induced by high glucose and its possible mechanism. Methods The RMCs proliferation induced by high glucose were observed by MTT assay and BrdU-ELISA as- say respectively. The cytotoxicity was detected by lactate dehydrogenase (LDH) assay. In ad- dition, the contents of malonaldehyde (MDA) and glutathione peroxidase (GSH) and the ac- tivities of superoxide dismutase (SOD) were measured using biochemical method. Results Carnosine significantly inhibited proliferation of RMCs induced by high glucose in concentra- tion-dependant way within 5 - 30 mmol/L. The inhibition of carnosine was closely related with its antioxidation. Conclusion Carnosine can inhibit proliferation of RMCs induced by high glucose through the mechanism of its antioxidation.
